凍干血小板聚集和活化的實(shí)驗(yàn)研究

【摘要】:目的了解血小板在凍干前、后功能活性的變化。方法應(yīng)用流式細(xì)胞儀檢測(cè)凍干復(fù)水(再水化)血小板及未冷凍干燥血小板胞膜CD61、CD62p、PAC-1的分子表達(dá),評(píng)價(jià)凍干復(fù)水后血小板活化狀態(tài);應(yīng)用血小板聚集儀檢測(cè)通過誘導(dǎo)劑瑞斯托霉素、二磷酸腺苷誘導(dǎo)的血小板最大聚集率,分析凍干血小板復(fù)水后血小板聚集率變化。結(jié)果血小板凍干保護(hù)劑滲透壓值為(2 055.6±123.8)m OSmol/kg;凍干前及復(fù)水后血小板分布寬度(PDW)(%)分別為15.50±3.05、18.29±0.55,MPV(f L)分別為9.01±1.84、8.63±0.58;凍干血小板復(fù)水回收率(%)為81.57±12.57;冷凍前及復(fù)水后血小板最大聚集率(%)分別為34.30±33.14、22.10±23.25;冷凍前及凍干復(fù)水后血小板胞膜CD61分子表達(dá)(%)分別為43.17±13.55、48.64±13.24,CD62p分子表達(dá)(%)分別為17.34±6.47、9.79±4.48,PAC-1分子表達(dá)(%)分別為4.92±6.32、4.22±4.64。結(jié)論冷凍前及復(fù)水后血小板最大聚集率的變化甚微,凍干復(fù)水后血小板仍具仍具有一定的存活能力;冷凍前及凍干復(fù)水后血小板胞膜CD61、CD62p和PAC-1分子表達(dá)略有變化(P0.05)。
[Abstract]:Objective to investigate the changes of platelet function before and after lyophilization. Methods flow cytometry was used to detect the molecular expression of CD61,CD62p,PAC-1 in the cell membrane of freeze-dried (rehydrated) platelets and non-freeze-dried platelets, and to evaluate the activation status of platelets after freeze-dried rehydration. The maximal platelet aggregation rate induced by the inducer ristoborubicin and adenosine diphosphate was measured by platelet aggregator and the change of platelet aggregation rate after rehydration of freeze-dried platelets was analyzed. Results the osmotic pressure of platelet freeze-dried protectant was (2 055.6 鹵123.8) m OSmol/kg;. The platelet distribution width (PDW) (%) before freeze-drying and after rehydration were 15.50 鹵3.05 鹵18.29 鹵0.55mv (f L), 9.01 鹵1.84 鹵8.63 鹵0.58, respectively, and the recovery rate of freeze-dried platelets was 81.57 鹵12.57, respectively. The maximum platelet aggregation rate (%) before freezing and after rehydration was 34.30 鹵33.140.10 鹵23.25, respectively. The expression of CD61 in platelets before freezing and after freeze-drying rehydration were 43.17 鹵13.55, 48.64 鹵13.24, CD62P and 17.34 鹵6.479.79 鹵4.48, respectively. The expression of PAC-1 (%) was 4.92 鹵6.32 鹵4.22 鹵4.64, respectively. Conclusion there is little change in platelet aggregation rate before freezing and after rehydration, and the platelet still has a certain viability after freeze-drying and rehydration. The expression of CD61,CD62p and PAC-1 in platelet membrane changed slightly before freezing and after freeze-dried rehydration (P0.05).
