【摘要】：目的:研究川芎嗪(ligustrazine,Lig)對內(nèi)毒素性休克大鼠腦損傷的作用并探討其作用機(jī)制。方法:將48只健康Wistar大鼠隨機(jī)分為正常組、LPS組和LPS+Lig組,每組16只,上述每組再分為2個亞組:6 h組和12 h組,各8只大鼠。以尾靜脈注射5 mg/kg脂多糖(LPS)建立內(nèi)毒素性休克大鼠模型,腹腔注射鹽酸川芎嗪注射液200 mg/kg,分別在相應(yīng)時點進(jìn)行眼球摘除采血并斷頭取腦組織,ELISA法檢測血清中神經(jīng)元特異性烯醇化酶(neuron-specific enolase,NSE)含量,腦組織勻漿檢測一氧化氮(nitric oxide,NO)含量,TUNEL染色法檢測腦組織中海馬區(qū)細(xì)胞凋亡情況,Western blot法檢測Bax和Bcl-2蛋白的表達(dá)。結(jié)果:與LPS組相比,川芎嗪能降低內(nèi)毒素性休克所致大鼠NSE和NO含量的上升,同時抑制海馬區(qū)細(xì)胞的凋亡,增加Bax的蛋白表達(dá)并降低Bcl-2的蛋白表達(dá)。結(jié)論:川芎嗪通過降低NSE和NO含量,減輕內(nèi)毒素性休克大鼠的腦損傷程度,可能與其調(diào)節(jié)Bax和Bcl-2蛋白的表達(dá)有關(guān)。
[Abstract]:Aim: to study the effect of ligustrazine Lig on brain injury in rats with endotoxic shock and its mechanism. Methods: Forty-eight healthy Wistar rats were randomly divided into normal group (n = 16) and LPS Lig group (n = 16). Each group was divided into 2 subgroups: 1: 6 h group and 12 h group, 8 rats in each group. Lipopolysaccharide (LPS) was injected into tail vein for 5 mg/kg to establish the rat model of endotoxic shock. Intraperitoneal injection of ligustrazine hydrochloride injection was performed at 200 mg / kg. Blood was taken from eyeball at the corresponding time point and brain tissue was taken from head off to detect the content of neuron-specific enolase in serum by Elisa. The content of nitric oxide (no) in brain homogenate and the expression of Bax and Bcl-2 protein in hippocampus were detected by Tunel staining and Western blot. Results: compared with LPS group, ligustrazine could decrease the contents of NSE and no, inhibit the apoptosis of hippocampal cells, increase the protein expression of Bax and decrease the protein expression of Bcl-2. Conclusion: Ligustrazine can decrease the level of NSE and no and reduce the degree of brain injury in rats with endotoxic shock, which may be related to its regulation of the expression of Bax and Bcl-2 proteins.
