氣相色譜質(zhì)譜聯(lián)用法對(duì)運(yùn)動(dòng)員尿液中內(nèi)源性類固醇水平特征的研究

【摘要】:目的:建立氣相色譜-質(zhì)譜聯(lián)用(GC-MS)同時(shí)測(cè)定尿液中6種內(nèi)源性類固醇激素水平的方法,對(duì)廣東省運(yùn)動(dòng)員體內(nèi)的類固醇激素代謝特征進(jìn)行統(tǒng)計(jì)歸納和比較分析,為廣東省運(yùn)動(dòng)員尿液中類固醇激素的生物護(hù)照的建立提供數(shù)據(jù)支持。方法:選取1 474名廣東省運(yùn)動(dòng)員(包括814名男隊(duì)員,660名女隊(duì)員)在賽前訓(xùn)練期間的尿樣,并對(duì)其中主要的6種內(nèi)源性類固醇進(jìn)行酶解、提取及衍生化后采用氣質(zhì)聯(lián)用(GC-MS)方法定量分析。分析采用HP-1MS色譜柱(17 m×0.2 mm,0.11μm),恒流模式,使用氦氣為載氣,流量為1 m L·min-1,進(jìn)樣量:1μL;分流比:10∶1;進(jìn)樣口溫度和EI離子源溫度分別為280℃和200℃;質(zhì)譜SCAN模式定性確證,掃描范圍m/z 40~460;選擇離子模式(SIM)下定量分析。結(jié)果:廣東省女運(yùn)動(dòng)員尿液中6種類固醇激素濃度及睪酮/表睪酮比值平均值均顯著低于國(guó)外女運(yùn)動(dòng)員(P0.05),其中睪酮濃度平均值僅為國(guó)外女運(yùn)動(dòng)員的12.2%;廣東省男運(yùn)動(dòng)員尿液中雄酮、本膽烷醇酮、睪酮、5β-雄烷-3α,17β-二醇4種類固醇激素濃度及睪酮/表睪酮比值平均值均顯著低于國(guó)外男運(yùn)動(dòng)員(P0.05),而表睪酮濃度平均值明顯高于國(guó)外男運(yùn)動(dòng)員(P0.05),5α-雄烷-3α,17β-二醇濃度平均值與國(guó)外男運(yùn)動(dòng)員基本持平(P0.05)。廣東省男、女運(yùn)動(dòng)員尿液中類固醇5α-雄烷-3α,17β-二醇/5β-雄烷-3α,17β-二醇濃度比值則均顯著高于國(guó)外男、女運(yùn)動(dòng)員的水平(P0.05),雄酮/本膽烷醇酮濃度比值與國(guó)外運(yùn)動(dòng)員基本持平。結(jié)論:本研究選取6種內(nèi)源性類固醇激素進(jìn)行氣質(zhì)聯(lián)用分析,并通過(guò)與國(guó)內(nèi)外運(yùn)動(dòng)員相關(guān)代謝指標(biāo)的對(duì)比分析,對(duì)廣東運(yùn)動(dòng)員訓(xùn)練期間的尿液中類固醇代謝特征進(jìn)行歸納,為廣東省運(yùn)動(dòng)員尿液內(nèi)源性類固醇指標(biāo)生物護(hù)照的建立提供數(shù)據(jù)支持。
