【摘要】:使用超高效液相色譜-串聯(lián)質(zhì)譜聯(lián)用技術(shù)(UPLC-MS/MS)建立了人尿中5種利尿劑的檢出方法。將尿樣用同體積的5%乙酸鉛水溶液沉淀蛋白質(zhì)并離心后,將上清液用多反應(yīng)監(jiān)測(MRM)模式進(jìn)行定性和定量分析。上述利尿劑1~5的檢測限(LOD)在0.5~2 ng/m L之間;線性范圍分別為3.0~700 ng/m L,1.5~1000 ng/m L,3.0~4000 ng/m L,6.0~5000 ng/m L和6.0~5000 ng/m L,線性良好(R20.99);在低、中和高3個(gè)濃度水平的日內(nèi)和日間RSD均小于15%。
[Abstract]:A method for the detection of five diuretics in human urine was established by UPLC-MS / MS. Urine samples were precipitated with 5% lead acetate solution of the same volume and centrifuged. The supernatants were qualitatively and quantitatively analyzed by multi-reaction monitoring (MRM) model. The detection limit (LOD) of diuretic 1D was between 0.5 and 2 ng/m / L, and the linear range was 3. 0 ~ 700 ng/m / L ~ 1. 5000 ng/m / L ~ 1. 5 000 ng/m / L ~ 3. 0 ~ 4 000 ng/m / L ~ 6. 0 ~ 5 000 ng/m / L and 6. 0 ~ 5 000 ng/m / L, respectively, with a good linearity (R 20. 99), and less than 1 515 ~ 5 000 ng/m / L within and between days at low, medium and high concentrations.
