【摘要】：目的了解深圳市無償獻(xiàn)血者乙肝病毒感染情況,分析其血清學(xué)和分子生物學(xué)特征。方法將收集到的確證HBs Ag(+)無償獻(xiàn)血者標(biāo)本分為/NAT(+)和NAT(-)兩組,進(jìn)行乙肝兩對(duì)半檢測(cè)以及病毒核酸大容量提取,采用巢氏PCR方法擴(kuò)增BCP/PC和S區(qū)基因序列,對(duì)所得的序列進(jìn)行分析。結(jié)果在42 297份血液中235份HBs Ag確證為陽性的標(biāo)本,HBs Ag陽性率為0.56%,其中NAT(+)162份,占總數(shù)68.9%,NAT(-)為73份,占31.1%,血清學(xué)模式以HBs Ag(+)、抗-HBe(+)、抗-HBc(+)為主,占69.78%,檢出10例抗-HBc(-)標(biāo)本,占總數(shù)的4.25%。在可測(cè)序166標(biāo)本中,B基因型所占88%(146/166),C基因型占11.4%(19/166),D基因型在HBs Ag(+)/NAT(+)出現(xiàn)1例。結(jié)論 NAT檢測(cè)與HBs Ag檢測(cè)方法均只能檢出部分乙肝感染獻(xiàn)血者,血液檢測(cè)必須結(jié)合兩種檢測(cè)方法來保證血液安全。數(shù)據(jù)顯示有必要應(yīng)用靈敏度更高的NAT檢測(cè)方法。
[Abstract]:Objective to investigate hepatitis B virus infection in blood donors in Shenzhen City and analyze its serological and molecular biological characteristics. Methods the samples of confirmed HBs Ag() donors were divided into two groups: nat () and NAT-). The HBV DNA was detected in two pairs and the virus nucleic acid was extracted in large volume. The nucleotide sequences of BCP/PC and S region were amplified by nested PCR method. The sequence was analyzed. Results the positive rate of HBs Ag in 235 of 42,297 blood samples confirmed to be positive for HBs Ag was 0.56. The positive rate of Nat (162 cases) was 73 (31.1%). The serological model was HBs AgA (HBs, anti-HBec, anti-HBcn (69.78g). Accounting for 4.25% of the total. Of the 166 samples sequenced, 88% of the alleles were genotype B (146R / 166C) and one case was found in HBs (Agna / NATA () with 11.4% / 166C / 166C genotype. Conclusion both NAT and HBs Ag detection methods can only detect some blood donors infected with hepatitis B, blood detection must be combined with two methods to ensure blood safety. The data show that it is necessary to apply a more sensitive NAT detection method.
