【摘要】:目的:觀察COBE Spectra血細(xì)胞分離機(jī)AutoPBSC程序以及Spectra Optia血細(xì)胞分離機(jī)MNC程序?qū)Σ杉慕】倒┱咄庵苎煅杉?xì)胞成分的影響。方法:2015年1月至2016年8月期間對12例供者隨機(jī)應(yīng)用COBE Spectra血細(xì)胞分離機(jī)AutoPBSC程序以及Spectra Optia血細(xì)胞分離機(jī)MNC程序進(jìn)行外周血造血干細(xì)胞采集,觀察應(yīng)用不同采集程序獲得的采集物中單個(gè)核細(xì)胞、CD34~+細(xì)胞、粒細(xì)胞、淋巴細(xì)胞、血小板數(shù)的差異。結(jié)果:兩種采集程序在循環(huán)血量、采集時(shí)間及抗凝劑使用量方面無顯著差異。COBE Spectra血細(xì)胞分離機(jī)AutoPBSC程序采集的采集物體積及單個(gè)核細(xì)胞計(jì)數(shù)均低于Spectra Optia血細(xì)胞分離機(jī)MNC程序的采集物體積及單個(gè)核細(xì)胞計(jì)數(shù)。AutoPBSC程序采集的CD34~+細(xì)胞計(jì)數(shù)高于MNC程序采集的CD34~+細(xì)胞計(jì)數(shù)。AutoPBSC程序采集物中混入的淋巴細(xì)胞及血小板較MNC程序中多,其差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:與Spectra Optia血細(xì)胞分離機(jī)MNC程序相比,COBE Spectra血細(xì)胞分離機(jī)AutoPBSC程序采集的造血干細(xì)胞數(shù)量多,混入的淋巴細(xì)胞及血小板多。
[Abstract]:Aim: to observe the effect of AutoPBSC procedure of COBE Spectra blood cell separator and MNC procedure of Spectra Optia blood cell separator on the components of peripheral blood hematopoietic stem cells collected from healthy donors. Methods: from January 2015 to August 2016, 12 donors were randomly collected from peripheral blood hematopoietic stem cells by AutoPBSC program of COBE Spectra blood cell separator and MNC program of Spectra Optia blood cell separator. The number of CD34 ~ cells, granulocytes, lymphocytes and platelets in mononuclear cells were observed. Results: two kinds of collecting procedures were in circulation blood volume, There was no significant difference in collection time and anticoagulant usage. The volume of samples and the count of mononuclear cells collected by AutoPBSC program of COBE Spectra blood cell separator were lower than those of MNC program of Spectra Optia blood cell separator, and the count of mononuclear cells was lower than that of Spectra Optia blood cell separator. The number of CD34 ~ cells collected by AutoPBSC program was higher than that by MNC program. The number of lymphocytes and platelets mixed in the collection of AutoPBSC program was more than that in MNC program. The difference was statistically significant (P 0.05). Conclusion: compared with the MNC procedure of Spectra Optia blood cell separator, the number of hematopoietic stem cells collected by AutoPBSC program is more than that of MNC procedure, and the number of lymphocytes and platelets mixed with AutoPBSC program is more than that of MNC procedure.
