【摘要】：目的:探討膿毒癥相關(guān)性腦病(sepsis-associated encephalopathy,SAE)大鼠海馬區(qū)神經(jīng)細(xì)胞自噬現(xiàn)象及微管相關(guān)蛋白1輕鏈3(LC3)的表達(dá)。方法:盲腸結(jié)扎穿孔術(shù)(cecal ligation and puncture,CLP)建立膿毒癥大鼠模型。60只30日齡健康雄性Wistar大鼠被隨機(jī)分為假手術(shù)組(10只)和CLP組(50只)。CLP后12 h監(jiān)測大鼠腦電圖(electroencephalogram,EEG)及體感誘發(fā)電位(somatosensory evoked potential,SEP),并進(jìn)行神經(jīng)生物學(xué)評分。根據(jù)大鼠是否發(fā)生SAE將CLP組大鼠再分為SAE(+)組和SAE( )組。HE染色觀察大鼠海馬區(qū)病理學(xué)改變;電子顯微鏡觀察大鼠海馬區(qū)神經(jīng)細(xì)胞自噬的超微結(jié)構(gòu);Western印跡檢測LC3-I和LC3-II蛋白的表達(dá)。結(jié)果:50只大鼠在CLP后12 h內(nèi)死亡5只,存活45只大鼠中有16只出現(xiàn)神經(jīng)行為學(xué)、EEG及SEP改變,診斷為SAE,其發(fā)病率為35.56%(16/45)。與假手術(shù)組和SAE( )組比較,SAE(+)組大鼠在CLP后12 h時(shí)α波的頻率明顯減少,δ波增加,P1振幅下降,P1波和N1波潛伏期延長(P0.05)。CLP后12 h時(shí)SAE(+)組大鼠海馬區(qū)細(xì)胞明顯水腫,錐體細(xì)胞明顯減少,甚至溶解,細(xì)胞排列紊亂,而假手術(shù)組和SAE( )組大鼠海馬區(qū)細(xì)胞形態(tài)正常、層次清楚。透射電子顯微鏡觀察表明:SAE(+)組大鼠海馬區(qū)細(xì)胞結(jié)構(gòu)紊亂,可見自噬泡、顆粒狀基質(zhì)和方形或長方形晶體,自噬泡內(nèi)含有溶酶體等細(xì)胞器,假手術(shù)組和SAE( )組大鼠海馬區(qū)組織細(xì)胞無自噬泡形成。SAE(+)組大鼠在CLP后12 h,海馬區(qū)神經(jīng)細(xì)胞LC3-II/LC3-I值明顯高于假手術(shù)組和SAE( )組(P0.05)。結(jié)論:SAE大鼠海馬區(qū)神經(jīng)細(xì)胞存在細(xì)胞自噬現(xiàn)象,LC3-II/LC3-I值明顯升高。
[Abstract]:Aim: to investigate the neuronal autophagy and the expression of microtubulosis-associated protein 1 light chain 3 (LC3) in the hippocampus of sepsis-associated encephalopathy (Sae) rats.Methods: sepsis rat model was established by cecal ligation and puncture). 60 male Wistar rats aged 30 days were randomly divided into sham operation group (n = 10) and CLP group (n = 50) 12 hours after cecal ligation and perforation, electroencephalogram (EEG) and somatosensory evoked electricity (SEG) were monitored in rats.Somatosensory evoked potentialSep, and neurobiological score.Rats in CLP group were subdivided into three groups according to whether SAE occurred or not. The pathological changes of hippocampus were observed by HE staining and the expression of LC3-I and LC3-II protein were detected by ultrastructure of autophagy of rat hippocampal neurons by electron microscope.Results five of the 50 rats died within 12 hours after CLP, and 16 of the 45 survived rats showed changes in neurobehavioral EEG and SEP, which were diagnosed as SAE. The incidence rate was 35.56% / 45%.Compared with the sham-operated group and the SAE group, the frequency of 偽 wave decreased significantly at 12 h after CLP, the amplitude of 未 wave increased and the latency of P1 decreased and the latency of N1 wave prolonged at 12 h after P0.05.
