微孔板稀釋法在活菌培養(yǎng)計(jì)數(shù)中的應(yīng)用觀察

【摘要】:目的以96孔板微量稀釋法代替?zhèn)鹘y(tǒng)試管稀釋法建立了新的活菌計(jì)數(shù)方法(也簡稱新方法或96孔板計(jì)數(shù)法),通過兩種活菌培養(yǎng)計(jì)數(shù)方法的比較,驗(yàn)證新方法的準(zhǔn)確性、快捷性。方法對(duì)新方法的不同吹吸次數(shù)進(jìn)行比較,選擇最佳的吹吸次數(shù)。用兩種活菌培養(yǎng)計(jì)數(shù)方法同時(shí)對(duì)同一管枯草桿菌黑色變種芽孢懸液進(jìn)行計(jì)數(shù),驗(yàn)證新方法的準(zhǔn)確性;不同實(shí)驗(yàn)技術(shù)人員同時(shí)對(duì)同一管枯草桿菌黑色變種芽孢懸液用96孔板進(jìn)行系列稀釋,用滴液計(jì)數(shù)法計(jì)數(shù),比較不同人員操作的誤差率;用兩種活菌培養(yǎng)計(jì)數(shù)方法對(duì)同一種消毒劑進(jìn)行中和劑鑒定試驗(yàn),驗(yàn)證新方法在消毒效果檢測中應(yīng)用的可行性。結(jié)果經(jīng)不同吹吸次數(shù)的誤差率比較,20次為最佳吹吸次數(shù);96孔板微量計(jì)數(shù)法對(duì)枯草桿菌黑色變種芽孢懸液計(jì)數(shù)結(jié)果與傳統(tǒng)方法計(jì)數(shù)結(jié)果一致;3名不同實(shí)驗(yàn)技術(shù)人員采用新方法同時(shí)對(duì)一管菌懸液計(jì)數(shù),3人之間的誤差率為1.69%,符合2002年版《消毒技術(shù)規(guī)范》要求,且誤差率遠(yuǎn)低于新入職實(shí)驗(yàn)技術(shù)人員用傳統(tǒng)方法對(duì)枯草桿菌黑色變種芽孢懸液活菌培養(yǎng)計(jì)數(shù)結(jié)果;在一種消毒劑的中和劑鑒定試驗(yàn)中,新方法與傳統(tǒng)方法活菌培養(yǎng)計(jì)數(shù)結(jié)果一致。結(jié)論新的活菌計(jì)數(shù)法計(jì)數(shù)結(jié)果與傳統(tǒng)活菌培養(yǎng)計(jì)數(shù)結(jié)果一致,且不同實(shí)驗(yàn)技術(shù)人員之間的誤差率很小,新的培養(yǎng)計(jì)數(shù)方法可以應(yīng)用到消毒劑懸液定量殺菌試驗(yàn)中,且采用新的培養(yǎng)計(jì)數(shù)方法更方便、快捷,節(jié)省時(shí)間,對(duì)實(shí)驗(yàn)人員技術(shù)要求不高,適合進(jìn)一步推廣。
[Abstract]:Objective to establish a new method for counting live bacteria by using 96 hole plate microdilution instead of traditional test tube dilution. The accuracy and rapidity of the new method were verified by comparing two methods of counting living bacteria.Methods the different blowing times of the new method were compared and the best suction times were selected.In order to verify the accuracy of the new method, the spore suspensions of Bacillus subtilis var. Niger were counted by two living bacteria culture counting methods at the same time.At the same time, the same tube Bacillus subtilis var. Niger spore suspension was diluted by 96 hole plates and counted by drop counting method to compare the error rate of different operators.The neutralizing agent identification test of the same disinfectant was carried out by two methods of living bacteria culture counting to verify the feasibility of applying the new method in the detection of disinfection effect.Results the error rate of different suction times was compared. The best suction times were 20 times. The results of counting spores suspension of Bacillus subtilis var. Niger by 96 hole plate microcount method were consistent with those of traditional method.Using the new method, the error rate between three persons counting a tube of bacteria suspension was 1.69, which was in accordance with the requirements of the 2002 edition of the Technical Specification for Disinfection.And the error rate was much lower than that of the traditional method of counting the live bacteria from Bacillus subtilis var. Niger by traditional methods. In the neutralizer identification test of a disinfectant,The result of the new method is consistent with that of the traditional method.Conclusion the result of the new counting method is consistent with that of the traditional living bacteria count method, and the error rate among different technicians is very small. The new method can be applied to the quantitative germicidal test of disinfectant suspensions.The new culture counting method is more convenient, quick and time saving, which is suitable for further popularization.
