【摘要】：目的對基于表面增強拉曼光譜檢測血清指紋譜的試驗條件進行優(yōu)化。方法以正常人血清為例,銀膠溶液為活性基底,分別檢測不同血清用量(2.5~500μl)、不同孵育時間(10~30 min)、不同孵育溫度(4℃、室溫、37℃)及不同血清處理方法(萃取、去蛋白)的增強拉曼信號。結(jié)果及結(jié)論血清用量不宜超過50μl,與增強基底材料的比例1∶1到5∶1均適宜;孵育時間在10~30 min均可;孵育溫度為4℃、室溫、37℃均可;血清直接與增強基底混合信號效果極強,進行萃取和去蛋白處理后,拉曼信號會減弱。
