有氧運動促進老年前期小鼠快縮型骨骼肌蛋白質合成代謝的研究

【摘要】:目的:探討中等強度有氧運動對老年前期小鼠快縮型骨骼肌蛋白質合成代謝的保護作用及其可能性機制。方法:20只13月齡雄性CD-1小鼠根據體重隨機納入對照組(C組)與有氧運動干預組(E組),運動組以自體重3%的負荷進行8周游泳訓練,每日訓練1次,每次45min,每周訓練6天。末次運動后24h處死小鼠(禁食17h),采用便攜式血糖儀檢測血糖,氨基酸分析儀檢測血漿游離氨基酸譜,酶聯免疫法檢測血清胰島素,BCA法進行蛋白定量,免疫印跡法檢測mTOR~(Ser2448)、4E-BP1~(Thr37/46)與p70S6K~(Thr389)磷酸化表達以及MHCⅡ蛋白表達。結果:與對照組(C組)小鼠相比,有氧運動干預組(E組)小鼠血漿游離氨基酸水平表現出下降趨勢,其中亮氨酸、谷氨酸與甘氨酸含量下降顯著(P0.01)。腓腸白肌蛋白質總量、mTOR~(Ser2448)、4E-BP1~(Thr37/46)、p70S6K~(Thr389)磷酸化表達與MHCⅡ蛋白表達均明顯升高,有顯著性意義(P0.01)。結論:中等強度有氧運動可以促進老年前期小鼠快縮型骨骼肌蛋白質合成,可能具有削弱衰老性肌肉萎縮發(fā)生發(fā)展的潛力,其作用機制可能涉及肌細胞內mTOR/p70S6K信號通路的活化。
[Abstract]:Objective: to investigate the protective effect and possible mechanism of moderate intensity aerobic exercise on protein synthesis and metabolism of fast contraction skeletal muscle in presenile mice.Methods Twenty 13-month-old male CD-1 mice were randomly divided into control group (C group) and aerobic exercise intervention group (group E). Exercise group was trained with 3% body weight for 8 weeks, once a day for 45 minutes, 6 days a week.The mice were sacrificed 24 hours after the last exercise (fasting for 17 h), blood glucose was detected by portable blood glucose analyzer, plasma free amino acid spectrum was detected by amino acid analyzer, and serum insulin was measured by enzyme linked immunosorbent assay (Elisa) for protein quantification.The phosphorylated expression and MHC 鈪,
