胰高血糖素樣肽-2對全腸外營養(yǎng)小鼠腸黏膜和肝功能的保護作用

【摘要】:目的:探討胰高血糖素樣肽-2(GLP-2)對全腸外營養(yǎng)(TPN)小鼠腸黏膜結構與肝功能的影響及相關機制。方法:將30只小鼠隨機分為三組,即自由進食組(Chow組)、TPN組和TPN+GLP-2組。小鼠經頸外靜脈置管2 d后,TPN組接受TPN液,TPN+GLP-2組除輸注等量上述營養(yǎng)液外,還經頸外靜脈注射60μg/d的GLP-2。第7天處死小鼠,取眼球靜脈血檢測肝功能,留取十二指腸、回腸末端和肝組織,檢測相關蛋白和mRNA表達,并觀察腸黏膜和肝組織的形態(tài)學改變。結果:7 d后,TPN組小鼠體重明顯下降,小腸絨毛高度與隱窩深度均劣于TPN+GLP-2組和Chow組(P0.05)。TPN組小鼠腸黏膜損傷較嚴重(P0.05),其血清肝功能指標水平較TPN+GLP-2組和Chow組高(P0.05),而清蛋白水平較其他兩組低(P0.05)。病理顯示TPN組小鼠肝細胞變性壞死,小腸微絨毛稀少,排列不齊,出現倒伏、脫落,上皮細胞線粒體破壞嚴重,內有大量空泡。TPN+GLP-2組腸PCNA蛋白表達較TPN組多(P0.05),而cleaved caspase-3表達較少(P0.05)。TPN+GLP-2組腸LGR-5 mRNA表達較TPN組多(P0.05),而肝GRP94 mRNA表達較低(P0.05)。結論:GLP-2可減輕TPN治療引起的小鼠腸黏膜損傷,對維護肝功能有一定的保護作用。
[Abstract]:Objective: to investigate the effects of glucagon like peptide-2 (GLP-2) on intestinal mucosal structure and liver function in TPN mice. Methods: thirty mice were randomly divided into three groups. The mice received TPN solution TPN GLP-2 for 2 days. In addition to the same amount of the above nutrient solution, the mice were given 60 渭 g / d GLP-2.The mice were killed on the 7th day. The blood of eyeball vein was taken to detect liver function, duodenum, ileum terminal and liver tissue, expression of related protein and mRNA were detected, and morphological changes of intestinal mucosa and liver tissue were observed. Results the weight of mice in TPN group decreased significantly after 7 days. The villus height and crypt depth of small intestine were lower than those of TPN GLP-2 group and Chow group (P0.05. TPN group). The serum liver function index level was higher than that of TPN GLP-2 group and Chow group, while the level of albumin was lower than that of other two groups. The pathological findings showed that TPN was higher than that in TPN GLP-2 group and Chow group. Group B mice hepatocyte degeneration and necrosis, Small intestinal microvilli are rare, irregular, lodging, shedding, and the mitochondria of epithelial cells are destroyed seriously. There were a large number of vacuoles. TPN GLP-2 group expressed more PCNA protein than TPN group, while cleaved caspase-3 expression was less in the group of cleaved GLP-2 than that in the group of TPN. The expression of LGR-5 mRNA was lower in group A than that in group TPN, but the expression of GRP94 mRNA in liver was lower than that in group A (P 0.05). Conclusion the expression of cleaved caspase-3 in the intestine of mice treated with TPN can be alleviated. It can protect liver function to some extent.
