【摘要】：目的:觀(guān)察丹紅注射液對(duì)膿毒癥大鼠心肌損傷的保護(hù)作用并探討其可能機(jī)制。方法:雄性Wistar大鼠分為6組:正常組、假手術(shù)組、模型6 h組、模型24 h組、丹紅6 h組和丹紅24 h組,每組各8只。盲腸結(jié)扎并穿孔(CLP)法建立膿毒癥大鼠動(dòng)物模型,觀(guān)察各組心肌組織超微結(jié)構(gòu)改變,心肌組織丙二醛(MDA)、超氧化物岐化酶(SOD)和高遷移率族蛋白1(HMGB1)含量的變化。結(jié)果:CLP術(shù)后,模型組大鼠心肌組織MDA含量、HMGB1蛋白合成進(jìn)行性升高、SOD活力則持續(xù)降低,與假手術(shù)組比較有差異(P0.05),丹紅組較模型組同時(shí)間點(diǎn)MDA、HMGB1及SOD均有改善,改善有統(tǒng)計(jì)學(xué)差異(P0.05);超微結(jié)構(gòu)示模型組大鼠心肌細(xì)胞線(xiàn)粒體腫脹,部分肌纖維稀疏,至24 h時(shí)腫脹加重,部分空泡變性;丹紅組心肌細(xì)胞損傷輕于模型組同時(shí)間點(diǎn)。結(jié)論:丹紅注射液對(duì)膿毒癥大鼠心肌損傷有一定程度的保護(hù)作用,其機(jī)制可能與清除氧自由基,降低HMGB1含量有關(guān)。
[Abstract]:Objective: to observe the protective effect of Danhong injection on myocardial injury in septic rats and explore its possible mechanism. Methods: male Wistar rats were divided into 6 groups: normal group, sham operation group, model group for 6 h, model group for 24 h. The sepsis rat model was established by cecal ligation and perforation with 8 rats in each group for 6 h and 24 h group respectively. The ultrastructure of myocardium was observed in each group. The changes of myocardial malondialdehyde (MDA), superoxide dismutase (SOD) and high mobility group protein (1HMGB1) were observed. Compared with sham-operated group, Danhong group improved MDAHMGB1 and SOD at the same time point and improved P0.05.The ultrastructure showed that myocardial mitochondria were swollen and some muscle fibers were sparse in the model group, and the swelling was aggravated at 24 h, and the changes of MDAHMGB1 and MDAHMGB1 in the model group were more severe than those in the control group. Partial vacuolar degeneration, myocardial cell damage in Danhong group was lighter than that in model group at the same time point. Conclusion: Danhong injection has some protective effect on myocardial injury in septic rats, and its mechanism may be related to scavenging oxygen free radicals and decreasing HMGB1 content.
