重組人促紅細(xì)胞生成素聯(lián)合骨髓間充質(zhì)干細(xì)胞移植對大鼠體外循環(huán)腎炎癥反應(yīng)的影響

【摘要】:目的促紅細(xì)胞生成素(erythropoietin,EPO)及骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMSCs)均對腎損傷炎癥反應(yīng)有保護(hù)和修復(fù)作用,但兩者聯(lián)合應(yīng)用保護(hù)CPB所致腎炎癥反應(yīng)少見報道。文中觀察重組人EPO聯(lián)合BMSCs移植對體外循環(huán)(cardiopulmonary bypass,CPB)大鼠腎炎癥反應(yīng)的影響。方法 40只雄性SD大鼠隨機數(shù)字表法分為5組(n=8):假手術(shù)組、CPB組、EPO組、BMSCs組、EPO+BMSCs組,假手術(shù)組只建立CPB模型,不行CPB,其余4組均行CPB,流量100 m L/(kg·min)轉(zhuǎn)流并維持1 h,轉(zhuǎn)流后BMSCs組經(jīng)頸靜脈輸注BMSCs1.5×106;EPO組頸靜脈注射EPO 3000IU/kg;EPO+BMSCs組EPO、BMSCs聯(lián)合應(yīng)用;CPB組和假手術(shù)組頸靜脈輸注相同容量的等滲鹽水。轉(zhuǎn)流結(jié)束后24 h麻醉處死實驗動物,檢測大鼠血清中肌酐(Cr)、尿素氮(BUN)含量;采用HE染色觀察腎組織形態(tài);ELISA法測定血清IL-6、IL-10水平,Western Blot檢測腎組織中TNF-α、IGF-1的表達(dá)。結(jié)果與假手術(shù)組比較,CPB組大鼠血清Cr、BUN、IL-6、IL-10和腎組織中TNF-α表達(dá)量均升高,而IGF-1表達(dá)下降,差異均有統(tǒng)計學(xué)意義(P0.05)。EPO、BMSCs和EPO+BMSCs組血清Cr、BUN水平、IL-6、腎組織中TNF-α均較CPB組下降,而IL-10、腎組織中IGF-1水平較CPB組升高(P0.05),EPO+BMSCs組血清Cr[(30.26±0.11)μmol/L]、BUN[(6.69±0.20)mmol/L]、IL-6[(401.7±6.8)pg/m L]水平低于EPO組[(42.76±2.24)μmol/L、(9.47±0.50)mmol/L、(533.7±7.0)pg/m L]和BMSCs組[(43.84±3.03)μmol/L、(8.80±0.32)mmol/L、(609.4±6.1)pg/m L],而EPO+BMSCs組IL-10水平[(1055.2±2.8)pg/m L]高于EPO組[(535.1±3.0)pg/m L]和BMSCs組[(544.7±2.6)pg/m L],差異均有統(tǒng)計學(xué)意義(P0.05)。結(jié)論 EPO和BMSCs對大鼠CPB腎損傷具有保護(hù)作用,效果好于單一使用EPO或BMSCs,減輕CPB對腎的炎癥反應(yīng)可能是其保護(hù)機制。
[Abstract]:Objective both erythropoietin erythropoietin (EPO) and bone marrow mesenchymal stem cells (BMSCs) can protect and repair the inflammatory response of renal injury. The effects of recombinant human EPO combined with BMSCs transplantation on renal inflammation in cardiopulmonary bypass (CPB) rats were studied. Methods 40 male SD rats were randomly divided into four groups. Five groups were divided into two groups: sham operation group (BMSCs group), BMSCs group (BMSCs group), EPO BMSCs group (P < 0.05), EPO BMSCs group (P < 0.05). CPB model was only established in sham-operation group. No, the other four groups were given CPB, flow rate 100ml / kg 路min) and maintained for 1 hour. After bypass, the BMSCs group was infused via jugular vein with EPO 3000IUP / kg EPO BMSCs group and the sham operation group was given the same volume of isoosmotic saline by jugular vein infusion. The experimental animals were anesthetized 24 hours after bypass. The contents of creatinine, bun and bun in serum of rats were determined. The levels of IL-6 and IL-10 in serum were detected by HE staining and the expression of TNF- 偽 -IGF-1 in renal tissue was detected by Western Blot. Results compared with sham-operated group, the expression of IL-10 in serum and TNF- 偽 in renal tissue in CPB group was increased, while the expression of TNF- 偽 in renal tissue was decreased. There were significant differences in serum Crnbun level and IL-6 level between P0.05, EPO-BMSCs and EPO BMSCs groups. TNF- 偽 in renal tissue was lower than that in CPB group. The levels of IL-10, IGF-1 in renal tissue were significantly higher than those in CPB group (30.26 鹵0.11) 渭 mol/L] bun [6.69 鹵0.20 mmol / L] IL-6 [401.7 鹵6.8 渭 mol / mL] in EPO group [42.76 鹵2.24 渭 mol / mL] and BMSCs group [43.84 鹵3.03] 渭 mol / L BMSCs group [43.84 鹵3.03 渭 mol / L] 渭 mol / L [6.69 鹵0.20 渭 mol / L], while the IL-10 level in EPO BMSCs group [1055.2 鹵2.8pgm] was significantly higher than that in EPO group [53.35 鹵7.0pgmL] and BMSCs group [43.84 鹵3.03] 渭 mol / L [43.84 鹵3.03] 渭 mol / L = 8.80 鹵0.32 渭 mol / L + 60 鹵6.1 鹵6.1pgm / L], while the IL-10 level in EPO BMSCs group [1055.2 鹵2.8pg / L] was significantly higher than that in EPO group [53.35g / L 鹵53.35g / L]. It has protective effect on CPB renal injury in rats. The effect is better than that of EPO or BMSCs alone, and the protective mechanism may be to alleviate the inflammatory response of CPB to kidney.
