去乙酰化酶Sirtuins在干細(xì)胞功能調(diào)節(jié)中的作用及前景

【摘要】:背景:氧化應(yīng)激嚴(yán)重制約著干細(xì)胞治療效能,而去乙酰化酶Sirtuin(SIRT)家族作為細(xì)胞內(nèi)重要的抗氧化酶在調(diào)節(jié)干細(xì)胞結(jié)構(gòu)和功能完整性中發(fā)揮重要作用,能有效降低干細(xì)胞損傷和增強(qiáng)干細(xì)胞生存能力。目的:綜述SIRT家族在干細(xì)胞中的調(diào)控作用及其機(jī)制,為SIRT家族在干細(xì)胞相關(guān)病理及生理學(xué)研究提供有益的參考。方法:由第一作者檢索1990至2015年Web of Science平臺,PubM ed數(shù)據(jù)庫,CNKI中國知網(wǎng)數(shù)據(jù)庫收錄的與SIRT和干細(xì)胞氧化應(yīng)激有關(guān)的文獻(xiàn)。中文檢索詞為"去乙;,干細(xì)胞,細(xì)胞代謝";英文檢索詞為"SIRT,stem cells,oxidative stress,molecular mechanisms"。共納入55篇文章進(jìn)行綜述。結(jié)果與結(jié)論:NAD+依賴的SIRT家族是機(jī)體組蛋白和其他蛋白去乙;闹匾阁w,在調(diào)節(jié)機(jī)體新陳代謝、基因組穩(wěn)定性、DNA損傷修復(fù)、染色質(zhì)重塑和應(yīng)激反應(yīng)中起到了關(guān)鍵的調(diào)節(jié)作用。隨著干細(xì)胞和SIRT家族研究的逐步深入,將為改善干細(xì)胞治療能效從而為臨床進(jìn)行干細(xì)胞移植治療提供更多的依據(jù)。
[Abstract]:Background: oxidative stress severely restricts the efficacy of stem cell therapy. The sirt family of deacetylase, as an important antioxidant enzyme in cells, plays an important role in regulating the structural and functional integrity of stem cells. It can effectively reduce the damage of stem cells and enhance the viability of stem cells. Objective: to review the regulatory role and mechanism of SIRT family in stem cells. Methods: the Web of Science platform was searched by the first author from 1990 to 2015. The Chinese key words are "deacetylase, stem cell, cell metabolism", which are related to oxidative stress of SIRT and stem cells. The key word was "SIRT stem cells stress". Molecular mechanisms ". A total of 55 articles are reviewed. Results and conclusions. The dependent SIRT family is an important enzyme for deacetylation of histone and other proteins. It plays a key role in regulating metabolism, genomic stability, DNA damage repair, chromatin remodeling and stress response. It will provide more evidence for improving the energy efficiency of stem cell therapy and for clinical stem cell transplantation.
