丙酮酸鈉改善紅細(xì)胞儲存損傷的初步探討

【摘要】:目的探討丙酮酸鈉(SP)對紅細(xì)胞儲存損傷的影響。方法戊巴比妥鈉腹腔注射麻醉小鼠,同時機械通氣,通過心臟穿刺取血,CPDA-1作為抗凝劑和儲存液,過濾去除白細(xì)胞后平分2組(n=8),SP組:添加SP濃縮液使儲存紅細(xì)胞中SP的終濃度為2.5 mmol/L;生理鹽水(NS)組:按相同體積比加入等量的生理鹽水。400 g離心15min,調(diào)整Hct為70%-75%,4℃儲存14 d后,用全自動血細(xì)胞計數(shù)儀測定儲存紅細(xì)胞的血常規(guī);分別用三磷酸腺苷(ATP)、丙二醛(MDA)測定試劑盒測定儲存紅細(xì)胞中ATP、MDA水平。結(jié)果儲存14 d時,2組血細(xì)胞計數(shù)各指標(biāo)無明顯差異(P0.05),且均在正常范圍。NS與SP組紅細(xì)胞中ATP含量(nmol/mg)為:2.46±0.19 vs 2.82±0.21(P0.05);MDA含量(nmol/mg)為15.45±0.33 vs 14.66±0.87(P0.05)。結(jié)論在紅細(xì)胞儲存過程中,SP明顯提高了儲存紅細(xì)胞ATP水平、降低了MDA水平,具有改善紅細(xì)胞儲存損傷的作用。
[Abstract]:Objective to investigate the effect of sodium pyruvate (SPN) on erythrocyte storage injury. Methods pentobarbital sodium was injected intraperitoneally to anesthetized mice and mechanical ventilation was used to collect blood by heart puncture. CPDA-1 was used as anticoagulant and storage liquid. After leucocyte removal, CPDA-1 was divided into two groups: the final concentration of SP in stored red blood cells was 2.5 mmol / L by adding SP concentrate.
