【摘要】：目的:采用液相色譜-動(dòng)態(tài)串聯(lián)質(zhì)譜技術(shù),建立和驗(yàn)證同時(shí)檢測(cè)人尿樣中120種刺激劑、利尿劑和其它禁用物質(zhì)的方法。方法:尿樣經(jīng)同等體積的5%醋酸鉛水溶液沉淀蛋白質(zhì)后,上清液用動(dòng)態(tài)多反應(yīng)監(jiān)測(cè)模式(d MRM)進(jìn)行檢測(cè),并比較了傳統(tǒng)MRM與動(dòng)態(tài)MRM在檢測(cè)靈敏度上的差異。結(jié)果:所篩查禁用物質(zhì)的檢測(cè)限均介于0.05~20 ng/ml之間,達(dá)到世界反興奮劑機(jī)構(gòu)對(duì)禁用物質(zhì)檢測(cè)能力的要求。在低、中和高三個(gè)濃度級(jí)別的日內(nèi)和日間精密度均小于20%,準(zhǔn)確度和基質(zhì)效應(yīng)在80%~120%之間。結(jié)論:該方法具有靈敏度高、準(zhǔn)確可靠和重現(xiàn)性好等特點(diǎn)。動(dòng)態(tài)MRM模式只掃描某個(gè)禁用物質(zhì)保留時(shí)間附近區(qū)域,駐留時(shí)間大大增加,其檢測(cè)靈敏度遠(yuǎn)高于傳統(tǒng)MRM模式,更適合于多種禁用物質(zhì)的同時(shí)檢測(cè)。
[Abstract]:Objective: to establish and verify the simultaneous determination of 120 stimulators in human urine by liquid chromatography-dynamic tandem mass spectrometry. Methods: urine samples were precipitated with the same volume of 5% lead acetate solution and the supernatant was detected by dynamic multi-reaction monitoring mode. The detection sensitivity of traditional MRM and dynamic MRM was compared. Results: the detection limit of prohibited substances for screening was between 0.05 ng/ml and 20 ng/ml. Meet the requirements of the World Anti-Doping Agency for the ability to detect banned substances. The intra-day and inter-day precision at low, medium and high levels of concentration is less than 20%. The accuracy and matrix effect were between 80% and 120%. Conclusion: this method has high sensitivity. Dynamic MRM only scans the area near the retention time of a banned substance, and the residence time increases greatly. The detection sensitivity of dynamic MRM mode is much higher than that of traditional MRM mode. It is more suitable for simultaneous detection of many banned substances.
