Bruker MS與VITEK-MS鑒定近平滑念珠菌復(fù)合體的性能對(duì)比

【摘要】:目的評(píng)估中國(guó)醫(yī)院侵襲性真菌監(jiān)測(cè)網(wǎng)(CHIF-NET)10和11收集的近平滑念珠菌復(fù)合體原始鑒定、Bruker MS與VITEK MS鑒定結(jié)果。方法經(jīng)中心實(shí)驗(yàn)室分子生物學(xué)方法復(fù)核鑒定確認(rèn),CHIF-NET 10和11從全國(guó)22所醫(yī)院共收集到近平滑念珠菌復(fù)合體352株,包括近平滑念珠菌305株,似平滑念珠菌36株,擬平滑念珠菌11株;以復(fù)核鑒定結(jié)果為金標(biāo)準(zhǔn),評(píng)價(jià)各單位原始常規(guī)方法鑒定結(jié)果,并評(píng)估Bruker MS和VITEK MS基質(zhì)輔助激光解吸電離飛行時(shí)間質(zhì)譜(MALDI-TOF MS)對(duì)近平滑念珠菌復(fù)合體的鑒定性能。結(jié)果與分子生物學(xué)鑒定結(jié)果相比,近平滑念珠菌的各醫(yī)院原始鑒定準(zhǔn)確率為90.8%,9.2%的菌株被API 20C或VITEK-2錯(cuò)誤鑒定為光滑念珠菌、季也蒙念珠菌等;常規(guī)方法無(wú)法鑒定似平滑念珠菌和擬平滑念珠菌;Bruker MS和VITEK MS對(duì)305株近平滑念珠菌鑒定準(zhǔn)確率分別達(dá)到了98.4%和97.7%;Bruker Biotyper 3.0數(shù)據(jù)庫(kù)中有似平滑念珠菌和擬平滑念珠菌參考圖譜,但鑒定準(zhǔn)確率僅分別為33.3%和27.3%;VITEK MS V2.0沒(méi)有兩種菌的參考圖譜而無(wú)法鑒定。結(jié)論該項(xiàng)研究率先在國(guó)內(nèi)評(píng)估了Bruker MS和VITEK MS鑒定近平滑念珠菌復(fù)合體的性能,證明二者對(duì)于近平滑念珠菌的鑒定是比較可靠的。
[Abstract]:Objective to evaluate the Chinese hospital invasive fungal monitoring network (CHIF-NET) original c.parapsilosis complex identification of 10 and 11 were collected, Bruker MS and VITEK MS. The identification results confirmation method by the Central Laboratory of molecular biology methods review identification, CHIF-NET 10 and 11 from 22 hospitals across the country were collected 352 strains of Candida parapsilosis complex, including nearly 305 strains of Candida parapsilosis, like Candida paropsilosis in 36 strains, 11 strains of Candida to smooth; to review identification results as the gold standard, the evaluation results of conventional methods of each unit of original identification, and evaluation of Bruker MS and VITEK MS matrix assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) on the performance appraisal of c.parapsilosis complex results. Compared with the results of molecular identification, the accuracy rate of each hospital c.parapsilosis original identification for 90.8%, 9.2% strains were identified as API 20C or VITEK-2 smooth read error Zhuhai bacteria, Candida guilliermondii; conventional methods can not be identified as Candida parapsilosis and Candida Bruker MS and quasi smooth; VITEK MS on the accuracy of identification of 305 strains of Candida parapsilosis respectively 98.4% and 97.7%; Bruker Biotyper 3 database like Candida parapsilosis and Candida parapsilosis Ni Ping reference map, but the identification accuracy rate respectively. 33.3% and 27.3%; VITEK MS V2.0 no reference map of two kinds of bacteria which could not be identified. Conclusion this study evaluated the performance of the first in the domestic Bruker MS and VITEK MS identification of Candida parapsilosis complex, proving the two identification for Candida parapsilosis is reliable.
