【摘要】：目的:應(yīng)用點(diǎn)免疫金滲濾法篩選適宜膠體金標(biāo)記的抗人IgG(以下簡(jiǎn)稱二抗),并運(yùn)用多個(gè)評(píng)價(jià)指標(biāo)進(jìn)行比較和分析。方法:選擇三個(gè)不同公司的二抗用膠體金標(biāo)記,根據(jù)棋盤滴定法確定最佳標(biāo)記條件;運(yùn)用點(diǎn)免疫金滲濾法對(duì)三種二抗的膠體金標(biāo)記后檢測(cè)效能進(jìn)行比較,采用包蟲病人和健康對(duì)照血清,用包蟲病特異性抗原檢測(cè)包蟲病患者血清中的特異性抗體,評(píng)價(jià)最優(yōu)的二抗,并與酶聯(lián)免疫吸附測(cè)定法進(jìn)行比較。結(jié)果:A、B、C三種二抗標(biāo)記的最佳標(biāo)記條件為:p H均為8.5,加入量分別為38.4、24、19.2μg/ml,綜合評(píng)價(jià)二抗B檢測(cè)效能最優(yōu)。將其與酶聯(lián)免疫吸附測(cè)定法檢測(cè)效能進(jìn)行比較,兩種方法檢測(cè)結(jié)果的Kappa=0.895(P0.05),吻合度較強(qiáng)。結(jié)論:應(yīng)用點(diǎn)免疫金滲濾法,以及本文提出的評(píng)價(jià)指標(biāo),能夠篩選出最佳二抗,對(duì)檢測(cè)試劑盒中二抗的選擇具有重要的參考價(jià)值。
[Abstract]:Objective: the application of dot immunogold filtration assay for screening of colloidal gold labeled anti human IgG (hereinafter referred to as the two anti), and the use of multiple evaluation indexes were compared and analyzed. Methods: three different companies with two anti colloidal gold, according to determine the optimal labeling conditions with chessboard titration; immune gold filtration assay to compare three kinds of colloidal gold labeled anti two note after detection efficiency, the patients with hydatid disease and healthy control serum, with specific antibody of echinococcosis specific antigen in sera of patients with hydatid disease detection, evaluation of optimal two antibody, and ELISA method were compared. Results: A B, C, the best labeling conditions three two anti markers: P H = 8.5, adding 38.4,24,19.2 g/ml respectively, two comprehensive evaluation of anti B detection efficiency. The optimal ELISA detection efficiency comparison, the test results of the two methods Kappa=0.895 (P0.05) has strong agreement. Conclusion: the application of dot immunogold filtration assay and the evaluation index proposed in this paper can screen the best two resistance, which has important reference value for the selection of the two antibody in the detection kit.
