羅非魚(yú)膠原肽中試車間設(shè)計(jì)與中試生產(chǎn)

【摘要】:膠原肽不僅分子量小易于人體消化吸收,而且具有獨(dú)特的生物活性,主要表現(xiàn)為抗高血壓、抗氧化、抗腫瘤等方面,因此在醫(yī)療、保健及美容領(lǐng)域具有良好的應(yīng)用前景。本研究立足于企業(yè)的需求,設(shè)計(jì)一個(gè)符合保健品生產(chǎn)要求的膠原肽GMP中試車間,并以羅非魚(yú)膠原蛋白胨為原料,結(jié)合生物酶解技術(shù)、膜分離技術(shù)等現(xiàn)代生物技術(shù)在該GMP中試車間中制備具有生物活性的膠原肽。首先,設(shè)計(jì)一個(gè)膠原肽GMP中試車間。根據(jù)羅非魚(yú)膠原肽的特點(diǎn)進(jìn)行中試工藝論證,并在此基礎(chǔ)上完成物料衡算、設(shè)備選型和水電用量估算等。該項(xiàng)目產(chǎn)量盈虧平衡點(diǎn)為49.3噸,經(jīng)營(yíng)安全率達(dá)52.1%,投資回收期為4.4年。在完成設(shè)計(jì)的基礎(chǔ)上,繪制車間平面分布圖、人流物流分布圖、設(shè)備流程圖和設(shè)備平面布置圖。其次,以羅非魚(yú)膠原蛋白胨為研究對(duì)象,采用不同酶解工藝,研究發(fā)現(xiàn)利用對(duì)蝦加工副產(chǎn)物制備所得蝦頭酶的最佳酶解條件為:加酶量0.5%,酶解溫度37℃,反應(yīng)p H為8.0,酶解時(shí)間為3 h。堿性蛋白酶的酶解條件為:加酶量為0.1%,酶解溫度45℃,反應(yīng)p H是9.0,酶解時(shí)間為3 h。同時(shí)對(duì)兩種酶解液進(jìn)行分子量測(cè)定,發(fā)現(xiàn)酶解液的分子量相近,主要分布在2 k Da以下。根據(jù)實(shí)驗(yàn)優(yōu)化后的酶解條件,進(jìn)行羅非魚(yú)膠原肽中試生產(chǎn)工藝探究,最終確立的工藝流程是:酶解、微濾、超濾、納濾、噴霧干燥、包裝。同時(shí),中試生產(chǎn)過(guò)程必須遵循GMP相關(guān)標(biāo)準(zhǔn),嚴(yán)格控制各個(gè)操作步驟的參數(shù)及衛(wèi)生條件要求。而中試生產(chǎn)膜分離過(guò)程中隨著時(shí)間的延長(zhǎng),膜污染增大,膜通量逐漸下降。最后,本文對(duì)羅非魚(yú)膠原肽中試產(chǎn)品進(jìn)行性質(zhì)分析。對(duì)中試膜分離產(chǎn)生的各組分進(jìn)行生物活性分析,蝦頭酶酶解制備的羅非魚(yú)膠原肽具有較高的ACE抑制活性(IC50=0.52mg/m L),而堿性蛋白酶制備的羅非魚(yú)膠原肽具有較高的抗氧化活性,其DPPH自由基清除率的IC50=5.7 mg/m L。這兩種蛋白酶制備的膠原肽各組分分子量相近,均分布在1 k Da以下。另外,文中對(duì)蝦頭酶制備的膠原肽中試產(chǎn)品進(jìn)行性質(zhì)分析。產(chǎn)品感官特點(diǎn)是白色、無(wú)味、無(wú)肉眼可見(jiàn)雜質(zhì)。在膠原肽的功能特性方面,其吸水性、持水性和溶解性較高,吸油性低。羅非魚(yú)膠原肽產(chǎn)品模擬胃腸液消化后ACE抑制活性稍有降低(IC50=0.85mg/m L),但其體外抗高血壓活性仍然較高。本文利用膠原肽GMP中試車間實(shí)現(xiàn)羅非魚(yú)膠原肽的中試生產(chǎn),對(duì)中試產(chǎn)品進(jìn)行相關(guān)性質(zhì)分析。這些研究為高生物活性羅非魚(yú)皮膠原肽的產(chǎn)業(yè)化生產(chǎn)奠定堅(jiān)實(shí)的基礎(chǔ),為羅非魚(yú)加工副產(chǎn)物的高效利用提供有益參考,也為相關(guān)功能性食品的開(kāi)發(fā)提供科學(xué)依據(jù)。
[Abstract]:Collagen peptide is not only easy to digest and absorb, but also has unique biological activity, such as anti-hypertension, anti-oxidation, anti-tumor and so on, so it has a good application prospect in medical, health care and beauty fields. Based on the demand of enterprises, a pilot workshop of collagen peptide GMP was designed to meet the requirements of health products production. The collagen peptone of tilapia was used as raw material, and the bioenzymatic hydrolysis technology was used. Membrane separation and other modern biotechnologies were used to prepare bioactive collagen peptides in the GMP pilot plant. First, a collagen peptide GMP pilot workshop was designed. According to the characteristics of collagen peptide of tilapia, the pilot-scale technological demonstration was carried out, and the material balance, equipment selection and the estimation of water and electricity consumption were completed on this basis. The profit and loss breakeven point of the project is 49.3 tons, the operating safety rate is 52.1 and the investment recovery period is 4.4 years. On the basis of completion of the design, draw workshop plane distribution map, flow distribution map, equipment flow chart and equipment layout plan.
