基于差分進(jìn)化算法的酵母培養(yǎng)在線自適應(yīng)控制

【摘要】:酵母培養(yǎng)中,若碳源添加不足則會(huì)抑制菌體生長(zhǎng),無(wú)法達(dá)到高密度;若碳源添加過(guò)量則會(huì)引起副產(chǎn)物乙醇的積累,導(dǎo)致細(xì)胞受到毒害,并降低碳源的利用效率。為解決這一矛盾,提出了一種基于差分進(jìn)化算法(differential evlolution algorithm,DE)的在線自適應(yīng)底物流加策略(DE-PID)并將其用于釀酒酵母和畢赤酵母的培養(yǎng)過(guò)程。在釀酒酵母培養(yǎng)中,利用DE-PID策略對(duì)呼吸商實(shí)施定值控制,培養(yǎng)34 h后的細(xì)胞密度達(dá)到52.5 g-DCW/L,比傳統(tǒng)底物流加策略(DO-Stat)下的相應(yīng)值提高91%。在畢赤酵母培養(yǎng)過(guò)程中,采用DE-PID策略分別對(duì)溶解氧濃度(DO)和乙醇濃度實(shí)施定值控制。在這兩個(gè)批次中,DO均能夠被控制在穩(wěn)定水平,乙醇濃度均被控制于2g/L以下,培養(yǎng)34 h后的細(xì)胞密度分別達(dá)到112.25和113.25 g-DCW/L。
[Abstract]:In yeast culture, if the carbon source is insufficient, the cell growth will be inhibited, and the high density can not be achieved. If the carbon source is added too much, it will lead to the accumulation of byproduct ethanol, which will lead to the cells being poisoned and reduce the carbon source utilization efficiency. To solve this problem, an online adaptive bottom logistics strategy named DE-PID-based differential evlolution algorithm (DED) was proposed and applied to the cultivation of Saccharomyces cerevisiae and Pichia pastoris. In the culture of Saccharomyces cerevisiae, the value of respiratory quotient was controlled by DE-PID strategy. The cell density reached 52.5g-DCW / L after 34 h culture, which was 91% higher than that under traditional bottom-logistics strategy (DO-Stat). In the process of Pichia pastoris cultivation, the DE-PID strategy was used to control the concentration of dissolved oxygen (DOO) and ethanol respectively. In these two batches, do could be controlled to a stable level, ethanol concentration was controlled below 2g/L, and the cell density reached 112.25 and 113.25 g DCW / L after 34 h culture, respectively.
