【摘要】:Connectivity map(CMap)是基于RNA芯片研究藥物作用機(jī)制和藥物重定位的技術(shù)與數(shù)據(jù)資源,主要的優(yōu)勢(shì)在于能夠高效地對(duì)全基因組上的基因表達(dá)做整體分析。相對(duì)于低通量的藥理學(xué)、毒理學(xué)方法以及色譜技術(shù),該技術(shù)能以全面、高通量、準(zhǔn)確等優(yōu)點(diǎn)廣泛應(yīng)用于藥物研究。尤其對(duì)由多種化合物共同作用發(fā)揮療效的中藥,CMap技術(shù)可以突破"藥物-靶標(biāo)"測(cè)定技術(shù)的單一性困難,對(duì)中藥的作用機(jī)制做綜合性分析,從分子水平上闡明中藥的藥物作用機(jī)制,有利于開(kāi)發(fā)自主知識(shí)產(chǎn)權(quán)的中藥,也為中藥獲取國(guó)際認(rèn)可奠定理論基礎(chǔ)。本文系統(tǒng)介紹CMap技術(shù)在中藥研究方面的應(yīng)用潛力,同時(shí)對(duì)該技術(shù)研究中藥的作用機(jī)制、藥物重定位及鑒別有效成分進(jìn)行評(píng)價(jià),還對(duì)推動(dòng)中藥走向國(guó)際市場(chǎng)做了具體分析與展望。
[Abstract]:Connectivity mapp is a technology and data resource based on RNA chip to study the mechanism of drug action and drug relocalization. Its main advantage lies in its ability to efficiently analyze gene expression in the whole genome, compared with low throughput pharmacology. Toxicological methods and chromatographic techniques that can be used in a comprehensive, high-throughput, The advantages of accuracy are widely used in drug research. In particular, the CMap technique, which has the effect of multiple compounds, can break through the difficulty of the "drug-target" determination technique, and make a comprehensive analysis of the mechanism of the action of traditional Chinese medicine. Elucidating the mechanism of drug action on molecular level is beneficial to the development of traditional Chinese medicine with independent intellectual property rights, and also lays a theoretical foundation for obtaining international recognition of Chinese medicine. This paper systematically introduces the application potential of CMap technology in the research of traditional Chinese medicine. At the same time, the mechanism, reorientation and identification of active components of traditional Chinese medicine were evaluated, and the specific analysis and prospect of promoting Chinese medicine to the international market were made.
