文章摘要:目的觀察三七總皂苷能否改善高脂誘導脂肪肝模型大鼠體內氧化應激及胰島素抵抗(insulinresistance,IR)。方法50只SD大鼠平均分為正常組、模型組、三七總皂苷大劑量組[160mg/(kg·d)]、三七總皂苷小劑量組[80mg/(kg·d)]及雙環(huán)醇組[100mg/(kg·d)],每組10只。正常組采用標準飼料喂養(yǎng),模型組及給藥組采用高脂飼料喂養(yǎng)。正常組和模型組給予蒸餾水灌胃,給藥組灌胃給藥4周。4周后測肥胖評定指數(shù)(Lee’S指數(shù))、體重;測肝濕重;檢測各組大鼠肝功能(ALT、AST);檢測空腹血糖(fastingbloodglucose,F(xiàn)BG)、空腹胰島素(fastinginsulin,F(xiàn)INS)水平,,計算胰島素敏感指數(shù)(insulinsensitiveidex,ISI)及胰島素抵抗指數(shù)(insulinresistanceindex,IRl);檢測血清TNF.僅、丙二醛(malond.ialdehyde,MDA)、總超氧化物歧化酶(totalsuperoxidedisumutase,T.SOD)、羥自由基(.OH)及血清總抗氧化能力(totalantioxidantcapacity,T—AOC)水平。肝臟石蠟切片HE染色觀察病理學改變。結果與正常組比較,模型組大鼠肝濕重、Lee’S指數(shù)均升高(P〈0.05),光鏡下肝臟出現(xiàn)明顯脂肪變性。與模型組比較,三七總皂苷及雙環(huán)醇治療可以明顯改善肝濕重、Lee’S指數(shù)(P〈0.05,P〈0.01)及ALT、AST等指標(P〈0.05,P〈0.01);減輕肝細胞脂肪沉積。模型組大鼠血清.OH、MDA、TNF-α.較正常組均顯著增高(P〈0.01),T—SOD及T-AOC活性降低(P〈0.01),且伴有IR。與模型組比較,經過三七總皂苷治療后,-OH、MDA、TNF-α濃度降低(P〈0.05,P〈0.01),而T—SOD及T-AOC的活性恢復(P〈0.05,P〈0.01),IR也得到明顯改善(P〈0.01)。結論三七總皂苷的抗氧化應急效應及改善IR效應是其對非酒精性脂肪肝療效的部分機制。
Abstr:Objective To investigate whether total saponins ofPanax notoginseng (tPNS) can a- meliorate oxidative stress and insulin resistance (IR) in the high fat induced nonalcoholic fatty liver dis- ease (NAFLD) rat model and to explore the relationship between oxidative stress and IR. Methods To- tally 50 healthy rats were randomly divided into the normal control group ( NC), the model group, the high dose tPNS group (at the daily dose of 160 mg/kg), the low dose tPNS group (at the daily dose of 80 mg/kg), and the bicyclol group (at the daily dose of 100 mg/kg), 10 in each group. Rats in the NC group were fed with standard forage. Those in the rest group were fed with high fat forage. Distilled water was given by gastrogavage to those in the NC group and the model group. Corresponding medication was performed for 4 weeks. Four weeks later Lee's index and body weight were measured. All rats were sacri- ficed to detect the wet weight of livers. Their sera was isolated and detected to calculate liver functions (serum ALT and AST levels). Levels of fasting blood glucose (FBG) and fasting insulin (FINS) were de- tected. Insulin sensitive index (ISI) and insulin resistance index (IRI) were calculated. Serum levels of TNF-eL and malondialdehyde (MDA), contents of total superoxide dismutase (T-SOD), hydroxy radical level (-OH), and total antioxidant capacity (T-AOC) were measured. Pathological changes of livers was observed by HE staining of paraffin section. Results Compared with the NC group, rats' wet liver weight and Lee's index increased in the model group (P 〈0.05), and results of light microscopy showed that ob- vious fatty degeneration occurred in livers.
