【摘要】：目的觀察復(fù)方芪鷹顆粒對糖尿病周圍神經(jīng)病變(DPN)大鼠坐骨神經(jīng)Hsp70表達(dá)及坐骨神經(jīng)損傷纖維的影響,探討其機制。方法從60只Wistar大鼠中隨機取出10只為正常組,50只大鼠造DPN模型,將成模43只大鼠隨機分為模型組(11只)、復(fù)方芪鷹顆粒低劑量組(11只)、復(fù)方芪鷹顆粒中劑量組(11只)、復(fù)方芪鷹顆粒高劑量組(10只),實驗過程中,模型組死亡3只,復(fù)方芪鷹顆粒低劑量組死亡1只。復(fù)方芪鷹顆粒低、中、高劑量組分別為1.17、2.34、4.68 g/kg,模型組、正常組均灌以相應(yīng)容積的蒸餾水,1次/d。治療4周后,觀察各組Hsp70表達(dá)、坐骨神經(jīng)損傷纖維變化。結(jié)果復(fù)方芪鷹顆粒各劑量組及模型組坐骨神經(jīng)中Hsp70表達(dá)與正常組比較,差異有高度統(tǒng)計學(xué)意義(P0.01);復(fù)方芪鷹顆粒中、高劑量組坐骨神經(jīng)中Hsp70表達(dá)與模型組比較,差異有高度統(tǒng)計學(xué)意義(P0.01),而中劑量及高劑量組間比較差異無統(tǒng)計學(xué)意義(P0.05);復(fù)方芪鷹顆粒各劑量組及模型組與正常組在在坐骨神經(jīng)損傷纖維百分比方面比較,差異有高度統(tǒng)計學(xué)意義(P0.01);復(fù)方芪鷹顆粒低、中、高劑量組與模型組在坐骨神經(jīng)損傷纖維百分比方面比較,差異有高度統(tǒng)計學(xué)意義(P0.01)。結(jié)論復(fù)方芪鷹顆�？赡芡ㄟ^調(diào)節(jié)DPN大鼠坐骨神經(jīng)Hsp70水平,從而改善受損坐骨神經(jīng)。
[Abstract]:Objective to observe the effect of compound Qiying granule on the expression of Hsp70 and injured fibers in sciatica of diabetic peripheral neuropathy (DPN) rats and to explore its mechanism. Methods 10 Wistar rats were randomly selected from 60 DPN rats as normal group and 50 rats as DPN model. 43 rats were randomly divided into model group (n = 11) and compound Qiying granule low dose group (n = 11), and the model group (n = 11) was randomly divided into model group (n = 11) and compound Qiying granule low dose group (n = 11). In the middle dose group (n = 11) and the high dose group (n = 10), 3 rats died in the model group and 1 in the low dose group. The low, middle and high dose groups of compound Qiying granule were 1.17, 2.34 and 4.68 g 路kg ~ (- 1), respectively. the model group and the normal group were treated with distilled water of the corresponding volume once a day. After 4 weeks of treatment, the expression of Hsp70 and the changes of nerve injured fibers in each group were observed. Results the expression of Hsp70 in the sciatica of each dose group and model group was significantly different from that of the normal group (P 0.01). Compared with the model group, the expression of Hsp70 in the sciatica of the high dose group was significantly different from that of the model group (P 0.01), but there was no significant difference between the middle dose group and the high dose group (P 0.05). Compared with the normal group, the percentage of fibers in each dose group and model group of compound Qiying granule was significantly different from that in the normal group (P 0.01). Compared with the model group, the percentage of fibers in the high dose group and the model group was significantly different from that in the model group (P 0.01), and the percentage of fibers in the high dose group was significantly higher than that in the model group (P 0.01). Conclusion compound Qiying granule may improve the injured sciatica by regulating the level of Hsp70 in the sciatica of DPN rats.
