針刺對糖尿病抑郁大鼠血糖、情緒晝夜節(jié)律及Per1mRNA、Per2mRNA表達的影響

【摘要】:目的:本實驗應(yīng)用行為學(xué)觀察、時間生物學(xué)研究方法及分子生物學(xué)技術(shù)等,研究糖尿病抑郁大鼠的血糖-HPA軸-情緒晝夜節(jié)律的改變特點,并且探討針刺對Per1mRNA、 Per2mRNA的基因表達的影響,揭示針刺調(diào)節(jié)糖尿病抑郁的血糖-HPA軸-情緒晝夜節(jié)律紊亂的相關(guān)分子生物機理。方法:將32只雄性SD大鼠,隨機分為空白組、糖尿病組、糖尿病抑郁組及針刺組,每組8只。空白組給予普通喂養(yǎng);糖尿病組給予高脂喂養(yǎng)及STZ試劑腹腔注射造模;糖尿病抑郁組給予高脂喂養(yǎng)、STZ試劑腹腔注射及慢性應(yīng)激造糖尿病抑郁模型;針刺組在糖尿病抑郁組造;A(chǔ)上針刺后三里、三陰交及百會穴,留針15分鐘,連續(xù)治療7天。針刺治療后,采用股動脈取血及斷頸取腦法取出各組大鼠的血清及下丘腦組織,采用酶聯(lián)免疫吸附法測定皮質(zhì)醇水平,采用實時定量PCR技術(shù)對大鼠SCN內(nèi)的的per2mRNA基因的表達量進行檢測。結(jié)果:1.糖尿病組血糖及情緒: (1)與空白組比較,糖尿病組大鼠的血糖值升高(P0.05),曠場實驗數(shù)據(jù)值下降(P0.05)。(2)與空白組比較,糖尿病組大鼠的24小時內(nèi)的血糖谷峰值差異大(P0.05);且其24小時內(nèi)的血糖值及曠場實驗數(shù)據(jù)的峰值出現(xiàn)時間延后。2.糖尿病抑郁組血糖及情緒: (1)與空白組大鼠及糖尿病組大鼠比較,糖尿病抑郁組大鼠的血糖值升高(P0.05),曠場實驗數(shù)據(jù)值下降(P0.05)。 (2)與空白組比較,糖尿病抑郁組大鼠24小時內(nèi)血糖的谷峰值差異大(P0.05),糖尿病抑郁組大鼠24小時內(nèi)的血糖峰值出現(xiàn)時間延后,其24小時內(nèi)的曠場實驗數(shù)據(jù)值改變趨勢與空白組大鼠相反。3.針刺組血糖及情緒: (1)與糖尿病抑郁組比較,針刺組大鼠的血糖值下降(P0.05),其曠場實驗數(shù)據(jù)值上升(P0.05)。 (2)針刺組大鼠的血糖谷峰值差異大(P0.05),針刺組大鼠血糖值的次高峰與次低谷的出現(xiàn)時間與空白組相同,其峰谷值出現(xiàn)時間有向空白組移位的趨勢,且10:00-22:00的最大值與最小值無統(tǒng)計學(xué)意義(P0.05);針刺組大鼠曠場實驗數(shù)據(jù)與空白組均呈現(xiàn)雙峰雙谷的節(jié)律變化。4.各組的皮質(zhì)醇水平:與空白組比較,糖尿病組大鼠、糖尿病抑郁組大鼠及針刺組大鼠的皮質(zhì)醇水平升高(P0.05);且三組的皮質(zhì)醇比較:糖尿病抑郁組糖尿病組針刺組(P0.05)。5.各組的Per1mRNA、Per2mRAN基因表達:與空白組比較,糖尿病組大鼠、糖尿病抑郁組大鼠及針刺組大鼠的Per2mRNA的表達量升高(P0.05);與糖尿病抑郁組及糖尿病組大鼠比較,針刺組大鼠的Per2mRNA的表達量下降;各組大鼠的Per1mRNA的表達量比較:空白組針刺組糖尿病組糖尿病抑郁組P0.05)。結(jié)論:1、針刺具有改善糖尿病抑郁大鼠的血糖、抑郁情緒及調(diào)節(jié)其血糖、情緒節(jié)律的作用。2、針刺具有降低糖尿病抑郁大鼠的皮質(zhì)醇水平的作用。3、針刺具有調(diào)節(jié)糖尿病抑郁大鼠的Per2mRNA的表達的作用;針刺可能是通過調(diào)節(jié)Per2mRNA基因的表達,從而調(diào)節(jié)血糖及情緒節(jié)律紊亂,以治療糖尿病抑郁的。
[Abstract]:Objective: to study the changes of blood glucose-HPA axis-mood circadian rhythm in diabetic depression rats by behavioral observation, time biology and molecular biology, and to explore the effect of acupuncture on Per1mRNA,. The effect of Per2mRNA gene expression on the molecular biological mechanism of acupuncture regulating blood glucose-HPA axis-emotional circadian rhythm disorder in diabetic depression was revealed. Methods: 32 male SD rats were randomly divided into blank group, diabetic depression group and acupuncture group with 8 rats in each group. The diabetic depression model was established by high fat feeding and intraabdominal injection of STZ reagent, high fat feeding, intraabdominal injection of STZ reagent and chronic stress in diabetic depression group. On the basis of modeling in diabetic depression group, acupuncture group was treated with Sanli, Sanyinjiao and Baihui points for 15 minutes for 7 days. After acupuncture treatment, the serum and inferior colliculus brain tissue of rats in each group were taken out by the method of taking blood from femoral artery and severing neck, and the level of cortisol was measured by enzyme-linked immunosorbent assay (Elisa). The expression of per2mRNA gene in rat SCN was detected by real-time quantitative PCR. Results: 1. Blood glucose and emotion in diabetic group: (1) compared with the blank group, the blood glucose value in the diabetic group was higher than that in the blank group (P 0.05), and the data value in the open field experiment (P 0.05). (2) was lower than that in the blank group. The peak value of blood glucose in diabetic group was significantly different within 24 hours (P 0.05). The peak value of blood glucose and open field experimental data within 24 hours was delayed. 2. Blood glucose and emotion in diabetic depression group: (1) compared with blank group and diabetic group, the blood glucose value of diabetic depression group was higher than that of blank group and diabetic group (P 0.05), and the data value of open field experiment was decreased (P 0.05). (2) compared with the blank group, the peak value of blood glucose in the diabetic depression group was significantly different from that in the blank group within 24 hours (P 0.05), and the peak value of blood glucose in the diabetic depression group was delayed within 24 hours. The change trend of open field experimental data within 24 hours was opposite to that of blank group. 3. Blood glucose and emotion in acupuncture group: (1) compared with diabetic depression group, the blood glucose value of acupuncture group decreased (P 0.05), and the data value of open field experiment increased (P 0.05). (2) the peak value of blood glucose in acupuncture group was significantly different (P 0.05). The secondary peak and subtrough of blood glucose value in acupuncture group were the same as those in blank group, and the appearance time of peak and valley value tended to shift to blank group.
