壯藥籬欄網(wǎng)質(zhì)量標準研究

【摘要】:目的:對壯藥籬欄網(wǎng)進行真?zhèn)、?yōu)劣評價,并初步建立壯藥籬欄網(wǎng)藥材質(zhì)量標準。方法:(1)通過基源鑒定、性狀鑒定、顯微鑒定與理化鑒定系統(tǒng)地對籬欄網(wǎng)進行生藥學定性研究。(2)照《中華人民共和國藥典》2015年版四部中規(guī)定的方法對籬欄網(wǎng)藥材進行限量檢查,包括水分、總灰分、酸不溶性灰分、浸出物等。(3)按照中藥化學預實驗方法對3個產(chǎn)地籬欄網(wǎng)進行成分預試。(4)以蘆丁為對照品,采用紫外-可見光分光光度法對廣西12個批次籬欄網(wǎng)藥材進行總黃酮含量測定,確定其所含總黃酮的限量。(5)以綠原酸為對照品,采用高效液相色譜法對廣西12個批次籬欄網(wǎng)藥材進行綠原酸含量測定,以確定其所含綠原酸的限量。(6)基于抗菌效價的生物檢定法檢測籬欄網(wǎng)藥材的生物活性。結(jié)果:(1)基源、性狀、顯微及薄層色譜鑒別均具顯著性特征,可作為鑒別籬欄網(wǎng)藥材真?zhèn)蔚囊罁?jù)。(2)實驗結(jié)果表明,籬欄網(wǎng)藥材水分含量范圍為6.26%~9.96%,初步擬定其水分不得超過11.00%;總灰分含量范圍為6.31~11.55%,初步擬定總灰分不得超過13.00%;酸不溶性灰分含量范圍為0.20%~1.60%,初步擬定酸不溶性灰分不得超過1.70%;浸出物含量在4.69%~9.22%之間,初步擬定醇溶性浸出物含量不得少于4.00%。(3)化學成分預實驗顯示籬欄網(wǎng)中含有黃酮類、糖、多糖、苷類、生物堿、蒽醌類、蛋白質(zhì)、氨基酸、有機酸及鞣質(zhì)或酚類等化學成分類型。(4)廣西12批次的籬欄網(wǎng)藥材總黃酮含量測得值在5.13%~14.32%之間,初步擬定其所含總黃酮不得少于4.00%。(5)廣西12批次籬欄網(wǎng)藥材中綠原酸含量測得值在0.22%~0.64%之間,初步擬定其所含綠原酸(C16H18O9)不得少于0.20%。(6)籬欄網(wǎng)藥材的對數(shù)劑量和抑菌效應呈線性關(guān)系,相關(guān)性好,基于抗菌效價的生物檢定法可行,初步擬定籬欄網(wǎng)(0.5g·ml-1)的效價不得低于0.8000U/ml。結(jié)論:為籬欄網(wǎng)藥材的準確鑒定和質(zhì)量標準的制定提供了科學依據(jù),也為籬欄網(wǎng)的開發(fā)應用研究奠定了基礎。
[Abstract]:Objective: to evaluate the authenticity, merits and demerits of Zhuang medicine fence net, and to establish the quality standard of Zhuang medicine fence net. Methods: (1) through base source identification, character identification, The pharmacognosy of hedgerow net was systematically studied by microscopic identification and physical and chemical identification. (2) according to the methods stipulated in the 2015 edition of the Pharmacopoeia of the people's Republic of China, the limited amount of medicinal materials in hedgerow net, including moisture and total ash, was examined. Acid insoluble ash, extract, etc. (3) according to the chemical pre-experimental method of traditional Chinese medicine, the composition of hedgerow net from three producing areas was pre-tested. (4) rutin was used as reference material. The content of total flavonoids in 12 batches of hedgerow in Guangxi was determined by UV-vis spectrophotometry, and the limit of total flavonoids was determined. (5) chlorogenic acid was used as reference material. The content of chlorogenic acid in 12 batches of hedgerow net in Guangxi was determined by high performance liquid chromatography (HPLC) to determine the limit of chlorogenic acid. (6) the biological activity of hedgerow net was determined by bioassay based on antibacterial titer. Results: (1) the identification of base source, character, microscope and thin layer chromatography was significant, which could be used as the basis for distinguishing the authenticity of hedgerow net. (2) the experimental results showed that the water content of hedgerow net was 6.26% and 9.96%, The moisture content of the preliminary formulation shall not exceed 11.00%; The range of total ash content is 6.31 鈮,
