黃芪多糖促進(jìn)脊髓損傷大鼠運(yùn)動(dòng)功能恢復(fù)的實(shí)驗(yàn)研究

目的本實(shí)驗(yàn)主要觀察黃芪多糖(Astragalus Polysaccharide;APS)對(duì)脊髓損傷(Spinal Cord Injury;SCI)大鼠運(yùn)動(dòng)功能恢復(fù)及組織病理變化的影響,并進(jìn)一步檢測(cè)黃芪多糖對(duì)脊髓損傷大鼠GAP-43(Growth-associated protein-43)及MAP2(Microtubule-associated protein 2)蛋白表達(dá)的影響,闡明其治療脊髓損傷的作用機(jī)制及靶點(diǎn),從而明確黃芪多糖對(duì)脊髓損傷的治療作用。方法1.黃芪多糖對(duì)大鼠脊髓損傷后運(yùn)動(dòng)功能障礙和脊髓病理?yè)p害的修復(fù)作用70只成年Sprague-Dawley大鼠,體重200-220g,隨機(jī)分為正常組(n=10)、損傷組(n=30)和黃芪多糖治療組(n=30),損傷組和黃芪多糖治療組內(nèi)部再分為7 d、14 d、28 d三個(gè)亞組。損傷組和治療組均采用Allen打擊法(10g×25 mm)制備大鼠T10脊髓損傷模型。造模成功后,治療組于術(shù)后腹腔注射黃芪多糖注射液(Sigma公司)10mg/Kg,每天一次,持續(xù)至取材為止。術(shù)后7 d、14 d、28 d,各亞組分別行BBB評(píng)分,HE染色觀察組織...
