川陳皮素通過調(diào)控RORα改善自然衰老小鼠骨流失(英文)

隨著世界人口老齡化的加速,老年性骨質(zhì)疏松癥的發(fā)病率呈現(xiàn)出明顯的上升趨勢(shì).衰老相關(guān)的骨質(zhì)流失的機(jī)制研究對(duì)尋找有效的治療骨質(zhì)疏松藥物具有重要意義.天然產(chǎn)物為治療骨質(zhì)疏松癥藥物的發(fā)現(xiàn)提供了豐富的來源.川陳皮素是柑橘屬植物果實(shí)中含量最豐富的黃酮類成分之一,具有多種生物活性.在本研究中以20個(gè)月自然衰老小鼠為模型,連續(xù)15天腹腔注射給予川陳皮素,Micro-CT結(jié)果表明川陳皮素可以增加自然衰老小鼠骨體積分?jǐn)?shù)和降低骨小梁分離度,改善自然衰老小鼠的骨微觀結(jié)構(gòu),HE染色結(jié)果表明川陳皮素可增加骨小梁表面成骨細(xì)胞數(shù)量,川陳皮素處理小鼠血清中骨鈣蛋白(Osteocalcin)水平顯著增加.在細(xì)胞水平,川陳皮素處理小鼠前成骨細(xì)胞(MC3T3-E1),Mki67水平顯著增加,骨堿性磷酸酶水平和骨鈣蛋白水平增加,茜素紅染色明顯增強(qiáng),表明川陳皮素能促進(jìn)MC3T3-E1細(xì)胞增殖、分化和礦化.進(jìn)一步機(jī)制研究發(fā)現(xiàn)川陳皮素可直接激活視黃酸受體相關(guān)孤兒受體α(retinoic acid receptor-related orphan receptors,RORα),并以RORα依賴的方式下調(diào)骨硬化蛋白(Sclerostin,...
