基于ADME的大黃素肝毒性及相關(guān)機(jī)制研究

一、研究背景與目的大黃素是存在于大黃和何首烏等藥材中的天然蒽醌類物質(zhì),具瀉下、抗菌、抗炎、抗腫瘤和保護(hù)肝腎等作用,因而受到了廣泛的關(guān)注和研究。然而大黃素對(duì)肝臟具雙重作用,小劑量起保護(hù)作用,大劑量產(chǎn)生毒性。目前關(guān)于大黃素代謝與毒性的關(guān)系尚不明確,本研究主要考察以下科學(xué)問(wèn)題:①大黃素的肝細(xì)胞毒性及細(xì)胞色素P450(Cytochromes P450,CYPs)介導(dǎo)的I相代謝對(duì)毒性的影響;②葡萄糖醛酸化轉(zhuǎn)移酶(UDP-glucuronosyltransferases,UGTs)介導(dǎo)的Ⅱ相代謝對(duì)毒性的影響及轉(zhuǎn)錄因子對(duì)關(guān)鍵UGT酶的調(diào)控;③大鼠長(zhǎng)期攝入大黃素致肝毒性具性別差異的機(jī)制;④丙磺舒合用大黃素對(duì)大鼠肝毒性的影響及機(jī)制。為大黃素的臨床合理使用提供一定的理論基礎(chǔ)和科學(xué)依據(jù)。二、研究方法1.采用HepG2細(xì)胞模型探討大黃素的肝細(xì)胞毒性,并采用CYP1A2/3A4的抑制劑和誘導(dǎo)劑,考察Ⅰ相代謝對(duì)毒性的影響。2.結(jié)合體外UGTs純酶實(shí)驗(yàn)、55例人肝/36例人腎樣本的基因組和轉(zhuǎn)錄組學(xué)數(shù)據(jù)探究參與大黃素Ⅱ相代謝的體內(nèi)主要UGT酶。分別敲低HepG2細(xì)胞中UGT2B7和轉(zhuǎn)錄因子HNF4A,探討Ⅱ相代謝與毒...

N-乙酰半胱氨酸在體內(nèi)可轉(zhuǎn)化為谷胱甘肽,補(bǔ)充細(xì)胞內(nèi)源性谷胱甘肽,增??強(qiáng)細(xì)胞解毒功能。采用0.5?mMN-乙酰半胱氨酸預(yù)處理HepG2細(xì)胞,1?h后再加??入大黃素,檢測(cè)各給藥組GSH水平和細(xì)胞生存率情況。如圖2-5所示,單獨(dú)加??入50?大黃素可降低GSH水平。將N-乙酰半胱氨酸單獨(dú)加入至細(xì)胞內(nèi),GSH??水平明顯增加。采用N-乙酰半胱氨酸預(yù)處理細(xì)胞,再加入大黃素后,GSH水平??雖高于對(duì)照組,但明顯低于單用N-乙酰半胱氨酸組,細(xì)胞生存率高于單用大黃??素組,這是由于大黃素消耗了由N-乙酰半胱氨酸轉(zhuǎn)化的谷胱甘肽,減少了對(duì)細(xì)??胞內(nèi)還原型谷胱甘肽的消耗,從而減輕了對(duì)細(xì)胞生長(zhǎng)的抑制作用。??A「n?GSH?B??料?方??p?丁?e丨仰-??卜?LJi?_??^?^j>c?N-acelylcysteine?〇?M):?^?^?^??^?^?v-**-?Fjnodin??19??
