振源膠囊對辛伐他汀及其活性代謝物在大鼠體內(nèi)的藥動(dòng)學(xué)影響(英文)

研究振源膠囊對辛伐他汀及其活性代謝產(chǎn)物辛伐他汀酸在大鼠體內(nèi)的藥動(dòng)學(xué)影響,為臨床兩藥聯(lián)合應(yīng)用提供一定的理論依據(jù)。將24只雄性Wistar大鼠隨機(jī)分成對照組與實(shí)驗(yàn)組,每組12只,對照組連續(xù)灌胃3‰羧甲基纖維素鈉15天,實(shí)驗(yàn)組連續(xù)灌胃振源膠囊混懸液15天。兩組大鼠于16日清晨均灌胃給予等量辛伐他汀5.3mg/kg。給藥后不同時(shí)間點(diǎn)進(jìn)行眼內(nèi)眥靜脈叢采血,離心即得血漿樣品。采用HPLC-MS/MS法測定辛伐他汀及辛伐他汀酸的血藥濃度。應(yīng)用DAS 2.1.1軟件擬合藥動(dòng)學(xué)參數(shù), SPSS 25.0軟件分析統(tǒng)計(jì)學(xué)差異。結(jié)果發(fā)現(xiàn),聯(lián)用振源膠囊后,辛伐他汀酸及辛伐他汀的藥動(dòng)學(xué)參數(shù)AUC0–24與Cmax均顯著性減少(P<0.05)。振源膠囊可能對CYP3A4酶有一定促進(jìn)作用,可減少辛伐他汀及其代謝產(chǎn)物在大鼠體內(nèi)的含量。
