pH敏感型仿病毒載藥系統(tǒng)的構(gòu)建與性能研究

近年來,受到病毒結(jié)構(gòu)和功能的啟發(fā),立足于從結(jié)構(gòu)仿生到功能仿生的角度,一系列模擬病毒組分和結(jié)構(gòu)的新型納米裝配體已被很好地構(gòu)建,并成功地提高了生物活性分子的遞送效率。透明質(zhì)酸(HA)是一類廣泛分布于細胞外基質(zhì)的聚陰離子多糖,研究發(fā)現(xiàn),大多數(shù)惡性腫瘤細胞表面過度表達研究發(fā)現(xiàn),大多數(shù)惡性腫瘤細胞表面過度表達HA的受體,如CD44,RHAMM和LYVE-1。值得注意的是,在各種惡性腫瘤組織中,透明質(zhì)酸酶(Hyaluronidase)的水平也是偏高的,因此HA很容易降解成低分子量的葡萄糖醛酸和N—乙酰氨基葡萄糖的雙糖重復單元片段。因此,HA不僅是載體的可降解保護層,還可作為配體靶向于特殊的細胞受體。在此基礎(chǔ)上本課題以透明質(zhì)酸為主體構(gòu)建了pH敏感的模擬病毒外殼的材料HA-Hyd-DOX,并合成了mPEG-Hyd-DOX作為對照。以這種材料構(gòu)建的仿病毒衣殼的聚合物膠束可有效的抵抗蛋白質(zhì)相互作用并延長血液循環(huán)時間。此外,HA-Hyd-DOX通過CD44受體介導的內(nèi)吞作用可以有效的被癌細胞攝取,而大大的減少成纖維細胞的攝取。同時腙鍵(Hyd)在溶酶體中pH=5的條件下可以被破壞,DOX被釋放出來從溶酶體中...
