福建沿海沉積物放線菌的多樣性及生物活性

【摘要】:目的探索福建沿海沉積物中可培養(yǎng)海洋放線菌的多樣性及其抗菌、抗腫瘤活性,為發(fā)現(xiàn)新的藥物先導(dǎo)化合物提供新菌源。方法從15份福建沿海沉積物選擇性分離放線菌;16S r RNA基因序列分析多樣性;采用10種活性篩選模型分析生物活性。結(jié)果共分離到216株海洋放線菌,MOPS-脯氨酸培養(yǎng)基的分離效果最好。選取77株代表菌株進(jìn)行16S r RNA基因序列分析,發(fā)現(xiàn)其歸屬于14個(gè)屬,其中10株為潛在新種。有164株(77%)海洋放線菌對(duì)8種指示菌中的一種或幾種具有抑制作用;有39株(18.1%)對(duì)He La細(xì)胞或Hep G2細(xì)胞有細(xì)胞毒活性(抑制率≥50%)。結(jié)論福建沿海沉積物樣品中含有豐富多樣的海洋放線菌,大量菌株具有抗菌與抗腫瘤活性,是天然產(chǎn)物開發(fā)的重要資源。
[Abstract]:Objective to explore the diversity, antibacterial and antitumor activities of cultured marine actinomycetes in coastal sediments of Fujian Province, and to provide a new source for the discovery of new drug lead compounds. Methods actinomycetes were selectively isolated from 15 coastal sediments of Fujian Province, the diversity of 16s r RNA gene sequence was analyzed, and 10 active screening models were used to analyze the biological activity. Results A total of 216 strains of marine actinomycetes were isolated, and the isolation effect of MOPS- proline medium was the best. 77 representative strains were selected for 16s r RNA gene sequence analysis.
