頂空氣相色譜法同時測定甲苯磺酸妥舒沙星原料藥中6種有機溶劑的殘留量

【摘要】:目的:建立同時測定甲苯磺酸妥舒沙星原料藥中甲醇、乙醇、二氯甲烷、正己烷、四氫呋喃和苯等有機溶劑殘留量的方法。方法:采用頂空氣相色譜法。色譜柱為以6%氰丙基苯基-94%二甲基聚硅氧烷(DB-624)為固定液的毛細(xì)管柱,程序升溫,進(jìn)樣口溫度為180℃,檢測器為氫火焰離子化檢測器,溫度為300℃,載氣為高純氮氣,載氣流速為1.5 m L/min,分流比為10∶1,頂空平衡溫度為100℃,平衡時間為40 min,頂空瓶裝樣量為10 m L,頂空進(jìn)樣量為1 m L。結(jié)果:甲醇、乙醇、二氯甲烷、正己烷、四氫呋喃和苯檢測質(zhì)量濃度線性范圍分別為178.3~1 782.7μg/m L(r=0.999 1)、301.2~3 012.1μg/m L(r=0.999 7)、33.81~338.10μg/m L(r=0.999 3)、18.02~180.22μg/m L(r=0.999 1)、43.26~432.58μg/m L(r=0.999 1)、0.126 8~1.268 1μg/m L(r=0.999 1);定量限分別為0.31、3.00、0.67、0.02、0.005、0.10μg/m L,檢測限分別為0.15、1.51、0.22、0.01、0.001、0.05μg/m L;精密度試驗的RSD≤3.1%;穩(wěn)定性、重復(fù)性試驗中甲醇、正己烷的RSD≤5%;加樣回收率分別為93.72%~102.20%(RSD=3.1%,n=9)、90.10%~101.79%(RSD=4.0%,n=9)、97.07%~103.11%(RSD=2.0%,n=9)、92.38%~103.83%(RSD=3.9%,n=9)、95.44%~103.62%(RSD=2.8%,n=9)、94.00%~104.73%(RSD=4.1%,n=9)。結(jié)論:該方法快速、靈敏、準(zhǔn)確,適用于甲苯磺酸妥舒沙星原料藥中甲醇、乙醇、二氯甲烷、正己烷、四氫呋喃和苯等有機溶劑殘留量的同時測定。
