鹽酸帕羅西汀對(duì)卵巢切除大鼠學(xué)習(xí)記憶能力及血清雌激素水平的影響

【摘要】:目的探討抗抑郁劑鹽酸帕羅西汀對(duì)卵巢切除大鼠學(xué)習(xí)記憶能力、血清雌激素水平及大鼠海馬雌激素受體表達(dá)的影響。方法采用SD雌鼠雙側(cè)卵巢切除(OVX)作為雌激素波動(dòng)引起的抑郁與認(rèn)知障礙模型,用Morris水迷宮實(shí)驗(yàn)來測(cè)試大鼠的學(xué)習(xí)記憶功能。實(shí)驗(yàn)共分為正常對(duì)照組、OVX對(duì)照組、OVX藥物組。予5-羥色胺重?cái)z取抑制劑類藥物:鹽酸帕羅西汀10 mg/(kg·d)干預(yù)4周。采用ELISA法測(cè)量SD大鼠血清黃體生成素、卵泡刺激素、雌激素。免疫組化法檢測(cè)大鼠海馬雌激素受體α(ERα)和雌激素受體β(ERβ)的陽性細(xì)胞表達(dá)數(shù)。結(jié)果 1 Morris水迷宮實(shí)驗(yàn)中,OVX對(duì)照組和OVX藥物組的逃避潛伏期明顯縮短,停留在平臺(tái)時(shí)間百分比明顯增加。2 OVX藥物組的血清雌激素較OVX對(duì)照組明顯增加(P0.05)。3藥物組與非藥物組的ERα、ERβ表達(dá)無明顯差異。結(jié)論帕羅西汀增加OVX大鼠血清雌激素水平和改善學(xué)習(xí)記憶能力,而對(duì)海馬ERα、ERβ表達(dá)無明顯影響。
[Abstract]:Objective to investigate the effects of paroxetine hydrochloric acid on learning and memory ability, serum estrogen level and estrogen receptor expression in hippocampus of ovariectomy rats. Methods bilateral ovariectomy (OVX) was used as a model of depression and cognitive impairment induced by estrogen fluctuation in SD female rats. Morris water maze test was used to test the learning and memory function of rats. The experiment was divided into normal control group, OVX control group and OVX drug group. 5-hydroxytryptamine reuptake inhibitor: paroxetine hydrochloric acid 10 mg/ (kg 路d) was given for 4 weeks. Serum luteinizing hormone, follicular stimulating hormone and estrogen were measured by ELISA method in SD rats. The expression of estrogen receptor 偽 (ER 偽) and estrogen receptor 尾 (ER 尾) in rat hippocampus was detected by immunohistochemistry. Results 1 in Morris water maze test, the escape latency of OVX control group and OVX drug group was significantly shortened and the percentage of staying on the platform was significantly increased. 2 the serum estrogen level in OVX drug group was significantly higher than that in OVX control group (P 0.05). 3 there was no significant difference in the expression of ER 偽 and ER 尾 between drug group and non-drug group. Conclusion Paroxetine increases serum estrogen level and improves learning and memory ability in OVX rats, but has no significant effect on the expression of ER 偽 and ER 尾 in hippocampus.
