高效液相色譜法測(cè)定喹紅霉素的有關(guān)物質(zhì)

【摘要】:目的建立高效液相色譜法測(cè)定喹紅霉素的有關(guān)物質(zhì)。方法采用Inertsil ODS-3(150mm×4.6mm,5μm)色譜柱,流動(dòng)相A為磷酸鹽緩沖液(0.04mol/L的磷酸氫二鉀溶液,20%的磷酸調(diào)節(jié)p H至5.8),流動(dòng)相B為乙腈,梯度洗脫;流速為1m L/min;檢測(cè)波長(zhǎng)為250nm;柱溫為35℃;進(jìn)樣體積為10μL。結(jié)果喹紅霉素與各有關(guān)物質(zhì)分離良好;在0.1007~40.28μg/m L濃度范圍內(nèi),峰面積與濃度呈良好的線性關(guān)系,相關(guān)系數(shù)為0.9999;檢測(cè)限為0.25ng,相當(dāng)于0.0025%雜質(zhì)量。結(jié)論該方法簡(jiǎn)便、靈敏、專屬、可靠,可用于喹紅霉素有關(guān)物質(zhì)的測(cè)定。
