【摘要】：目的應(yīng)用高效液相色譜技術(shù)建立鹽酸右哌甲酯緩釋微丸有關(guān)物質(zhì)檢查和含量測(cè)定的方法。方法采用Agilent ZORBAX SB-C18色譜柱(250 mm×4.6 mm,5μm);磷酸二氫鉀溶液[25 mmol/L,三乙胺0.2%(v/v),磷酸調(diào)至p H 3.5]-甲醇(65∶35)為流動(dòng)相,檢測(cè)波長(zhǎng)209 nm,流速1.0 ml/min,柱溫40℃。結(jié)果在選定的色譜條件下,鹽酸右哌甲酯與相關(guān)雜質(zhì)分離良好,制劑輔料不干擾鹽酸右哌甲酯的測(cè)定;檢測(cè)限為81.12 ng/ml(S/N=3),在質(zhì)量濃度2~30μg/ml范圍內(nèi),鹽酸右哌甲酯與峰面積線性關(guān)系良好,相關(guān)系數(shù)r為0.9995;平均回收率為100.83%,溶液在12 h內(nèi)保持穩(wěn)定(RSD為0.10%)。結(jié)論該方法簡(jiǎn)便、準(zhǔn)確、專屬性強(qiáng),可用于鹽酸右哌甲酯緩釋微丸有關(guān)物質(zhì)及其含量的測(cè)定。
