左氧氟沙星在肺炎克雷伯菌肺部感染小鼠體內(nèi)的藥動(dòng)學(xué)

【摘要】:目的:建立肺炎克雷伯菌(Klebsiella pneumoniae,Kp)肺部感染小鼠模型,采用高效液相色譜(HPLC)法測(cè)定不同劑量左氧氟沙星(levofloxacin,LVF)在其體內(nèi)的藥動(dòng)學(xué)參數(shù)。方法:血漿樣品經(jīng)乙腈沉淀蛋白后,采用Diamosail-C18色譜柱分離,流動(dòng)相為乙腈(0.05 mol·L~(-1))-磷酸二氫鉀(17∶83),磷酸二氫鉀鹽溶液以磷酸調(diào)整pH為3.0左右,流速為1 mL·min~(-1);檢測(cè)波長(zhǎng)為295 nm。結(jié)果:LVF在0.3~12 mg·L~(-1)內(nèi)線性關(guān)系良好;方法的日間和日內(nèi)精密度均小于7.2%,穩(wěn)定性良好。Kp肺部感染小鼠口服LVF15,30,60和90 mg·kg~(-1)后,LVF的PK參數(shù)tmax、t1/2、CL/F和Vd/F均無顯著性差異(P0.05)。結(jié)論:所建立的HPLC分析方法準(zhǔn)確、靈敏、專屬性強(qiáng)、重復(fù)性高,適用于肺炎克雷伯菌肺部感染小鼠體內(nèi)的左氧氟沙星藥動(dòng)學(xué)研究。

The pharmacokinetics parameters of levofloxacin (levofloxacin,LVF) in vivo were determined by high performance liquid chromatography (HPLC). Methods: the plasma sample was precipitated by acetonitrile and separated by Diamosail-C18 column. The mobile phase was acetonitrile (0.05 mol 路L ~ (- 1)-potassium dihydrogen phosphate (17:83). The pH of potassium dihydrogen phosphate solution was adjusted to about 3.0 with phosphate, and the flow rate was 1 mL 路min ~ (- 1). Detection wavelength is 295 nm.
