丙烯腈誘發(fā)活性氧生成對(duì)大鼠精子質(zhì)量及超微結(jié)構(gòu)的影響

【摘要】:目的探討丙烯腈(ACN)誘導(dǎo)氧化應(yīng)激對(duì)大鼠附睪精子質(zhì)量與鞭毛超微結(jié)構(gòu)的影響。方法 40只雄性SPF級(jí)健康SD大鼠按體重隨機(jī)分為4組,染毒組以12.5、25和50 mg/kg ACN灌胃,對(duì)照組給予等體積玉米油。1次/d,6 d/周,連續(xù)13周。測精子活性氧(reactive oxygen species,ROS)、丙二醛(methane dicarboxylic aldehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、精子線粒體膜電位(mitochondrial membrane potential,MMP)和精子質(zhì)量(0.86%生理鹽水中游離精子),電子顯微鏡下觀察精子鞭毛超微結(jié)構(gòu)。結(jié)果 50 mg/kg組精子MMP較其他組均降低(P0.05)。50 mg/kg組精子ROS含量較其他組升高(P0.05)。染毒組SOD活力較對(duì)照組降低(P0.05)。精子鞭毛超微結(jié)構(gòu)顯示,25和50 mg/kg ACN組精子鞭毛外部致密纖維和線粒體結(jié)構(gòu)有損傷。精子密度50 mg/kg組為(3.52±0.46)×10~6/ml,較對(duì)照組、12.5和25 mg/kg組[分別為(5.71±0.66)×10~6/ml、(4.95±1.33)×106/ml和(5.99±0.78)×106/ml]均降低(P0.05);25和50 mg/kg組精子活率分別為(62.18±3.4)%和(66.25±7.23)%,較對(duì)照組(83.22±13.86)%降低(P0.05)。d級(jí)精子率50 mg/kg組為(40.76±1.47)%,25 mg/kg組為(37.77±3.41)%,與對(duì)照組(27.77±4.85)%和12.5 mg/kg組(31.77±4.85)%比較均升高(P0.05)。精子平均路徑速度對(duì)照組高于各ACN組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。直線速度50 mg/kg組較對(duì)照組降低(P0.05)。精子ROS含量與精子MMP呈負(fù)相關(guān)(r=-0.643,P=0.024),與SOD活力呈負(fù)相關(guān)(r=-0.713,P=0.009),與精子活力呈負(fù)相關(guān)(r=-0.629,P=0.028);與精子平均路徑速度呈負(fù)相關(guān)(r=-0.839,P=0.001);與d級(jí)精子率呈正相關(guān)(r=0.699,P=0.011)。結(jié)論 ACN可致附睪內(nèi)精子ROS含量增高,精子鞭毛內(nèi)線粒體和外部致密纖維超微結(jié)構(gòu)改變,而使精子活動(dòng)能力和運(yùn)動(dòng)速度降低。
[Abstract]:Objective to investigate the effect of oxidative stress induced by acrylic (ACN) on sperm quality and flagellum ultrastructure in rat epididymis. Methods 40 male SPF healthy SD rats were randomly divided into 4 groups according to body weight. The exposed group was given 12.5, 25 and 50 mg/kg ACN intragastrically, while the control group was given equal volume corn oil once a day for 6 days for 13 weeks. Sperm reactive oxygen species (reactive oxygen species,ROS), malondialdehyde (methane dicarboxylic aldehyde,MDA), SOD (superoxide dismutase,SOD), sperm mitochondrial membrane potential (mitochondrial membrane potential,MMP) and sperm mass (0.86% saline free sperm) were measured. The ultrastructure of sperm flagellum was observed under electron microscope. Results the sperm MMP in 50 mg/kg group was lower than that in other groups (P 0.05). The sperm ROS content in 50 mg/kg group was higher than that in other groups (P 0.05). The activity of SOD in the exposed group was lower than that in the control group (P 0.05). The ultrastructure of sperm flagellum showed that the outer dense fibers and mitochondria of sperm flagellum were damaged in 25 and 50 mg/kg ACN groups.
