【摘要】：目的建立風(fēng)濕免疫病患者服用甲氨蝶呤的液相色譜-質(zhì)譜聯(lián)用血藥濃度測(cè)定方法。方法采用甲醇沉淀蛋白,以氨基蝶呤作為內(nèi)標(biāo),用Epic C18色譜柱(4.6 mm×50 mm,3μm),甲醇(含0.1%甲酸)-水(含0.1%甲酸)為流動(dòng)相,梯度洗脫,流速0.6 m L·min~(-1),柱溫40℃。用電噴霧離子化源,正離子模式,多反應(yīng)監(jiān)測(cè)(MRM)掃描方式進(jìn)行監(jiān)測(cè)�？疾旆椒ǖ膶傩浴�(biāo)準(zhǔn)曲線、定量下限、精密度、準(zhǔn)確度、回收率、基質(zhì)效應(yīng)和穩(wěn)定性。結(jié)果甲氨蝶呤在0.5~200.0 ng·m L~(-1)范圍內(nèi)線性良好(r=0.999 2),定量下限為0.5ng·m L~(-1),日內(nèi)、日間精密度及準(zhǔn)確度RSD15%,回收率為76.80%~85.49%。結(jié)論該檢測(cè)方法快速、靈敏,滿足對(duì)服用甲氨蝶呤72 h內(nèi)的風(fēng)濕免疫病患者血藥濃度的準(zhǔn)確測(cè)定。
