Chiralpak AD和Chiralcel OD-H柱分離莨菪烷衍生物對映體的研究

【摘要】:目的采用正相手性色譜柱建立莨菪烷衍生物A和B對映體的高效液相色譜法(HPLC)手性分離方法。方法在正相條件下分別使用Chiralpak AD和Chiralcel OD-H手性柱,考察流動相組成、流速和柱溫對對映體手性分離的影響;計算對映體與固定相相互作用的焓變差值和熵變差值等熱力學參數(shù)。結(jié)果化合物A在Chiralpak AD柱上得到最佳色譜分離條件;衔顱在Chiralcel OD-H柱上得到最佳色譜分離條件。在15℃~35℃柱溫范圍內(nèi),lnα對1/T呈線性關系,其手性分離過程受焓控制。結(jié)論此HPLC手性分離方法可用于化合物A和B的光學純度分析和鑒定。
[Abstract]:Objective to establish a high performance liquid chromatography (HPLC) (HPLC) chiral separation method for enantiomers of scopolane derivatives A and B by normal phase chiral column. Methods Chiralpak AD and Chiralcel OD-H chiral columns were used to investigate the effects of mobile phase composition, flow rate and column temperature on the chiral separation of enantiomers. The thermodynamic parameters such as enthalpy change difference and entropy change difference of enantiomer and stationary phase interaction are calculated. Results the optimum chromatographic separation conditions of compound A were obtained on Chiralpak AD column.
