【摘要】：目的:比較雷貝拉唑鈉右旋體、左旋體和消旋體對(duì)H+,K+-ATP酶活性抑制作用的強(qiáng)弱,以及體內(nèi)抗實(shí)驗(yàn)性大鼠胃潰瘍作用的強(qiáng)弱。方法:取正常SD大鼠胃黏膜,體外測(cè)定雷貝拉唑鈉右旋體、左旋體以及消旋體對(duì)H+,K+-ATP酶的抑制作用。建立水應(yīng)激型、消炎痛刺激、幽門(mén)結(jié)扎法3種大鼠胃潰瘍模型,觀察三者對(duì)實(shí)驗(yàn)性胃潰瘍形成的影響。并在水應(yīng)激模型中取SD大鼠胃黏膜測(cè)定體內(nèi)對(duì)H+,K+-ATP酶活性的影響。結(jié)果:右旋體、左旋體以及消旋體體外抑制H+,K+-ATP酶的活性未見(jiàn)明顯差異;但在水應(yīng)激模型中體內(nèi)對(duì)H+,K+-ATP酶活性有抑制作用,且雷貝拉唑鈉右旋體左旋體和消旋體。與模型組比較,雷貝拉唑鈉右旋體對(duì)3種SD大鼠實(shí)驗(yàn)性胃潰瘍具有明顯的抑制作用(P0.01或P0.05),效果優(yōu)于消旋體和左旋體,能顯著抑制胃潰瘍的發(fā)生。結(jié)論:體外H+,K+-ATP酶活性抑制測(cè)定結(jié)果,三者之間抑制效果沒(méi)有明顯差異;但體內(nèi)酶活性抑制作用雷貝拉唑鈉右旋體強(qiáng)于消旋體和左旋體。等劑量雷貝拉唑鈉右旋體對(duì)3種大鼠胃潰瘍的抗?jié)冏饔脧?qiáng)于消旋體和左旋體。
[Abstract]:Aim: to compare the inhibitory effects of Rabeprazole sodium on the activity of H, K-ATP and the anti-gastric ulcer in rats. Methods: the gastric mucosa of normal SD rats was taken to determine the inhibitory effects of Rabeprazole sodium dextrose, levoscele and racemate on H, K-ATP enzyme in vitro. The models of gastric ulcer induced by water stress, indomethacin and pyloric ligation were established in rats, and the effects of these three methods on the formation of experimental gastric ulcer were observed. The effect of SD rat gastric mucosa on the activity of H, K-ATP enzyme was measured in water stress model. Results: there was no significant difference in the activity of H, K-ATP between right-handed, left-handed and racemic bodies in vitro, but the activity of H, K-ATP was inhibited in water stress model in vivo, and the left-hand and racemic bodies of Rabeprazole sodium dextrose and racemate were found to have inhibitory effect on the activity of H, K-ATP in water stress model. Compared with the model group, Rabeprazole sodium dextrose had obvious inhibitory effect on experimental gastric ulcer in three SD rats (P0.01 or P0.05), which was better than racemic body and left-handed body, and could significantly inhibit the occurrence of gastric ulcer. Conclusion: there is no significant difference in inhibition effect of H, K-ATP enzyme activity among them in vitro, but the inhibitory effect of Rabeprazole sodium dextrose in vivo is stronger than racemates and levosomes. The anti-ulcer effect of ribeprazole sodium dextrose on three kinds of gastric ulcer in rats was stronger than that of racemic body and left-handed body in the same dose of Rabeprazole sodium.
