微透析-高效液相色譜法聯(lián)用研究吳茱萸提取物的經(jīng)皮吸收藥動(dòng)學(xué)特性

【摘要】:目的:利用在體皮膚微透析技術(shù)研究吳茱萸提取物經(jīng)皮吸收的特性。方法:以裸鼠為研究對(duì)象,建立在體經(jīng)皮微透析采樣技術(shù),以吳茱萸堿和吳茱萸次堿為指標(biāo)成分,采用高效液相色譜測(cè)定吳茱萸提取物經(jīng)皮給藥后透析液中的藥物濃度,通過(guò)相對(duì)損失率的校正,計(jì)算皮膚藥物濃度,并利用Kinetica 5.0軟件對(duì)皮膚藥物濃度和時(shí)間進(jìn)行非房室模型擬合,計(jì)算相關(guān)統(tǒng)計(jì)參數(shù)。結(jié)果:吳茱萸提取物中吳茱萸堿的達(dá)峰時(shí)間為(150±15.3)min,半衰期(t1/2)(263.7±41.6)min;吳茱萸次堿的達(dá)峰時(shí)間為(90±2.1)min,半衰期(t1/2)(194±17.3)min;二者均能較快地達(dá)到峰值,并在較長(zhǎng)的時(shí)間內(nèi)保持穩(wěn)定釋放,使皮下組織中的藥物濃度保持在一個(gè)相對(duì)恒定的水平。結(jié)論:本研究中所建立的微透析方法可用于吳茱萸提取物的皮膚藥動(dòng)學(xué)研究,吳茱萸堿和吳茱萸次堿可透過(guò)皮膚吸收而發(fā)揮臨床藥效。
[Abstract]:Objective: to study the characteristics of percutaneous absorption of Evodia officinalis extract by in vivo skin microdialysis. Methods: the in vivo percutaneous microdialysis sampling technique was established in nude mice. The concentration of Evodia officinalis extract in dialysate solution was determined by high performance liquid chromatography (HPLC) using evodipine and Evodia officinalis as the index components, and the drug concentration was determined by high performance liquid chromatography (HPLC) after transdermal administration of Evodia officinalis extract. The skin drug concentration was calculated by the correction of relative loss rate, and the non-atrioventricular model was used to fit the skin drug concentration and time with Kinetica 5.0 software, and the relevant statistical parameters were calculated. Results: the peak time of evodiamine was (150 鹵15.3) min, (263.7 鹵41.6) min;. The peak time of evodipine was (90 鹵2.1) min,. (194 鹵17.3) min;. Both of them could reach the peak value quickly and keep stable release for a long time so that the drug concentration in subcutaneous tissue was kept at a relatively constant level. Conclusion: the microdialysis method established in this study can be used to study the skin pharmacokinetics of Evodia officinalis extract. Evodipine and evodipine can be absorbed through the skin to exert the clinical efficacy.
