刺五加內(nèi)生放線菌Streptomyces tauricus CWJ-107活性次級(jí)代謝產(chǎn)物研究

【摘要】:目的從藥用植物刺五加內(nèi)生放線菌Streptomyces tauricus CWJ-107次級(jí)代謝產(chǎn)物中發(fā)現(xiàn)抗耐甲氧西林金黃色葡萄球菌(MRSA)活性良好的天然產(chǎn)物。方法采用16S r RNA基因序列分析對(duì)菌株CWJ-107進(jìn)行初步分類鑒定,大量發(fā)酵獲得粗提物,通過(guò)硅膠柱層析、LH20凝膠柱層析、薄層色譜以及高效液相色譜等方法,以抗MRSA活性導(dǎo)向進(jìn)行目標(biāo)化合物的分離純化,利用核磁共振和質(zhì)譜鑒定化合物結(jié)構(gòu),并利用微孔板法測(cè)定化合物最小抑菌濃度(minimal inhibitory concentration,MIC),對(duì)其抗MRSA活性進(jìn)行評(píng)價(jià)。結(jié)果菌種鑒定結(jié)果顯示菌株CWJ-107為Streptomyces tauricus,從菌株CWJ-107的菌絲體中分離得到1個(gè)抗MRSA活性化合物107-A,經(jīng)波譜解析證實(shí)其結(jié)構(gòu)為糖基化蒽醌類化合物cinerubin B,抗菌活性測(cè)定結(jié)果證實(shí)其具有良好的抗MRSA活性,MIC值為1μg/m L。結(jié)論從刺五加內(nèi)生放線菌CWJ-107的代謝產(chǎn)物中分離到的107-A經(jīng)結(jié)構(gòu)鑒定為糖基化蒽醌類化合物cinerubin B,除此之外,本文首次報(bào)道了該化合物的抗MRSA活性。
[Abstract]:Objective to find a natural product with good (MRSA) activity against methicillin-resistant Staphylococcus aureus from the secondary metabolites of endophytic actinomycetes Streptomyces tauricus CWJ-107 of Acanthopanax senticosus (Acanthopanax senticosus). Methods 16s r RNA gene sequence analysis was used to classify and identify the strain CWJ-107, and a large number of crude extracts were obtained by fermentation. The crude extracts were obtained by silica gel column chromatography, LH20 gel column chromatography, thin layer chromatography and high performance liquid chromatography. The target compounds were separated and purified by MRSA activity orientation. The structures of the compounds were identified by NMR and MS, and the minimum inhibitory concentration (minimal inhibitory concentration,MIC) of the compounds was determined by microporous plate method. The anti-MRSA activity of the compounds was evaluated. Results the strain CWJ-107 was isolated from the mycelia of CWJ-107 by Streptomyces tauricus, and a compound of anti-MRSA activity 107-A was isolated from the mycelia of CWJ-107. The structure of the compound was identified by spectral analysis as cinerubin B, a glycosylanthraquinone compound. The results of antimicrobial activity showed that it had a good MRSA activity, and the MIC value was 1 渭 g / m 路L ~ (-1) 路m ~ (-1) 路L ~ (-1). Conclusion 107-A isolated from the metabolites of actinomyces senticosus CWJ-107 was identified as cinerubin B by structure. In addition, the anti-MRSA activity of the compound was reported for the first time.
