多西他賽囊袋張力環(huán)的加速釋放試驗方法及制備研究

【摘要】:目的考察多西他賽囊袋張力環(huán)加速釋放度試驗條件,并優(yōu)化多西他賽囊袋張力環(huán)的處方工藝。方法通過考察釋放介質(zhì)中乙醇含量(0%、5%、10%)和不同溫度(37和45℃)對多西他賽囊袋張力環(huán)釋藥速度的影響,建立制劑體外加速釋放度試驗條件。利用加速釋放度試驗方法考察多西他賽囊袋張力環(huán)的體外釋放度,篩選最優(yōu)處方。結(jié)果在介質(zhì)為含5%乙醇pH 7.4磷酸鹽緩沖溶液、溫度為(45±0.5)℃的加速釋放條件下,藥物釋放速度提高了4倍,加速釋放度與長期釋放度用二項式擬合最好(y=0.004 6x~2+0.437 4x+9.683 7,r~2=0.998 4);載藥量為30%,載體為PLGA5050(60K-100K=1∶1,W/W)的處方釋藥平穩(wěn),可持續(xù)釋藥30 d,符合Peppas equation釋藥模型。結(jié)論多西他賽囊袋張力環(huán)的釋藥特性可用加速釋放度試驗快速考察,該制劑體外釋藥平穩(wěn)緩慢,具有臨床應(yīng)用前景。
