熒光探針?lè)y(cè)定奧沙利鉑的方法及機(jī)理研究

【摘要】:目的:建立一種簡(jiǎn)單、快速、靈敏的巴馬汀熒光探針?lè)?用于測(cè)定抗癌藥物奧沙利鉑。方法:以?shī)W沙利鉑和巴馬汀探針對(duì)葫蘆[7]脲空腔競(jìng)爭(zhēng)為基礎(chǔ),巴馬汀和葫蘆[7]脲的水溶液本身無(wú)熒光,二者混合后體系熒光顯著增強(qiáng),但當(dāng)加入無(wú)熒光的奧沙利鉑后,體系的熒光又顯著猝滅。結(jié)果:在0.05~1.75μg·m L~(-1)的范圍內(nèi)奧沙利鉑的濃度與熒光猝滅值呈良好的線性關(guān)系,最低定量限為20 ng·m L~(-1),檢測(cè)限為6 ng·m L~(-1)。并通過(guò)核磁共振和密度泛函理論對(duì)其作用機(jī)理進(jìn)行了探究,證實(shí)奧沙利鉑與葫蘆[7]脲可以形成1∶1的穩(wěn)定包合物。結(jié)論:該方法可用于血漿中奧沙利鉑的測(cè)定。
[Abstract]:Objective: to establish a simple, rapid and sensitive fluorescence probe method for the determination of oxaliplatin. Methods: based on the competition of oxaliplatin and palmatine probe to the cavity of gourd [7] urea, there was no fluorescence in the aqueous solution of Bamatin and gourd [7] urea. The fluorescence of the system was also significantly quenched. Results: there was a good linear relationship between the concentration of oxaliplatin and fluorescence quenching in the range of 0.05 渭 g mL ~ (-1). The lowest quantitative limit was 20 ng mL ~ (-1) and the detection limit was 6 ng mL ~ (-1). The mechanism of its action was studied by NMR and density functional theory. It was proved that oxaliplatin and cucurbitu [7] urea could form a stable inclusion complex of 1:1. Conclusion: this method can be used for the determination of oxaliplatin in plasma.
