【摘要】：目的:制備碘化鉀微乳并考察其透皮吸收的效果。方法:以偽三元相圖為基礎,依據(jù)微乳區(qū)域大小,確定微乳的油相、乳化劑、助乳化劑和后二者的比例(Km),初步篩選微乳處方;采用改進的Franz擴散池和離體小鼠皮膚進行體外經(jīng)皮滲透試驗,比較所制5%、10%碘化鉀微乳與5%碘化鉀溶液單位面積的累積滲透量(Qn),并對微乳進行加速穩(wěn)定性試驗。結(jié)果:碘化鉀微乳的最優(yōu)處方為碘化鉀10%(m/V)、水23%、混合乳化劑(月桂山梨坦-乙醇Km值為1∶1)61.6%、油相(丙二醇單辛酸酯)15.4%;所制微乳的滲透過程符合一級速率方程,3種樣品的Q24 h分別為(5.40±0.28)、(8.75±0.42)、(2.12±0.20)mg/cm2;40℃下貯存4周后所制微乳與貯存前比較p H、含量、外觀均未見明顯變化。結(jié)論:成功制得具有透皮吸收作用的碘化鉀微乳。
[Abstract]:Objective: to prepare potassium iodide microemulsion and evaluate its transdermal absorption. Methods: on the basis of pseudo-ternary phase diagram and according to the size of microemulsion, the oil phase, emulsifier, assistant emulsifier and the ratio of co-emulsifier and the latter were determined by (Km),. The modified Franz diffusion cell and mouse skin were used for in vitro percutaneous osmotic test. The cumulative permeation capacity (Qn),) of 10% potassium iodide microemulsion and 5% potassium iodide solution per unit area was compared and the accelerated stability test of the microemulsion was carried out. Results: the optimum formulation of potassium iodide microemulsion was 10% (m / V) of potassium iodide, 23% water, 61.6% mixed emulsifier (Km = 1:1) and 15.4% oil phase (propylene glycol monooctanoate). The osmotic process of the prepared microemulsion was in accordance with the first-order rate equation.
