【摘要】：目的探討香煙煙霧暴露對大鼠睪丸結(jié)構(gòu)及睪丸組織雄激素結(jié)合蛋白(ABP)mRNA及蛋白表達(dá)水平的影響。方法清潔級成年雄性SD大鼠160只,隨機(jī)分為對照組和低(10支)、中(20支)、高(30支)劑量染毒組,各組大鼠分別染毒2、4、6、8、12周,共16組,每組10只。實(shí)驗(yàn)組采用呼吸道靜式染毒,每日1次,每次30 min。觀察大鼠睪丸組織結(jié)構(gòu),采用RT-PCR及Western Blot法檢測睪丸組織ABP mRNA及蛋白表達(dá)水平。結(jié)果染毒組大鼠睪丸病理切片可見,生精細(xì)胞排列紊亂疏松,支持細(xì)胞呈現(xiàn)空泡化。大鼠睪丸組織ABP mRNA和蛋白表達(dá)水平均隨染毒劑量的增加而降低,染毒4周及12周后中、高劑量組ABP基因mRNA表達(dá)水平低于對照組,染毒6周后高劑量組ABP基因mRNA表達(dá)水平低于對照組,染毒4~8周后高劑量組ABP蛋白表達(dá)水平低于對照組,染毒12周后低、中、高劑量組ABP蛋白表達(dá)水平低于對照組,差異均有統(tǒng)計學(xué)意義(P0.05)。結(jié)論香煙煙霧暴露可引起雄性大鼠睪丸組織受損,且睪丸組織ABP基因mRNA及蛋白的表達(dá)水平下降。
[Abstract]:Objective to investigate the effects of cigarette smoke exposure on the expression of androgen binding protein (ABP) mRNA and protein in testis of rats. Methods 160 clean grade adult male SD rats were randomly divided into control group (n = 10), medium (n = 20) and high dose (n = 30). The experimental group was treated with respiratory tract administration once a day for 30 min.. The structure of rat testis was observed and the expression of ABP mRNA and protein in testis was detected by RT-PCR and Western Blot. Results in the exposed group, testicular sections were observed, spermatogenic cells were disordered and loose, and Sertoli cells were vacuolated. The expression of ABP mRNA and protein in testis of rats decreased with the increase of exposure dose. After 4 and 12 weeks of exposure, the mRNA expression of ABP gene in high dose group was lower than that in control group. The expression level of ABP gene mRNA in high dose group was lower than that in control group after 6 weeks of exposure, the ABP protein expression level in high dose group was lower than that in control group after 8 weeks of exposure, and the expression level of ABP protein in middle and high dose group was lower than that in control group after 12 weeks of exposure. The difference was statistically significant (P0.05). Conclusion cigarette smoke exposure can damage the testis of male rats and decrease the expression of ABP gene mRNA and protein in testis.
