三氯異氰尿酸對斑馬魚胚胎及幼魚的發(fā)育毒性

【摘要】:目的探討新型消毒劑三氯異氰尿酸(TCCA)對斑馬魚胚胎及幼魚的發(fā)育毒性。方法選擇受精后2 h的斑馬魚胚胎進(jìn)行染毒:①將胚胎置于含有TCCA 50,100,150,200,250和300 mg·L~(-1)的培養(yǎng)液中暴露48 h,檢測半數(shù)致死濃度(LC_(50))。②將胚胎置于含有TCCA 0,10.4,20.8和41.7 mg·L~(-1)的培養(yǎng)液中暴露96 h,分別于暴露后48,72和96 h檢測胚胎死亡率、畸形率和心率,采用羥胺法于暴露后2,4和6 h檢測胚胎組織超氧化物歧化酶(SOD)的活性,HE染色觀察暴露后7 d幼魚頭部組織病理結(jié)構(gòu)。結(jié)果 TCCA暴露48 h,斑馬魚胚胎LC_(50)為166.9 mg·L~(-1)。與正常對照組相比,暴露后96 h,TCCA 41.7 mg·L~(-1)組斑馬魚胚胎死亡率和畸形率顯著升高(P0.05),TCCA各劑量組心率顯著下降(P0.05);暴露后72h,TCCA41.7mg·L~(-1)組死亡率顯著升高(P0.05),20.8和41.7mg·L~(-1)組畸形率顯著升高(P0.05),心率顯著下降(P0.05);暴露后48 h,TCCA 41.7 mg·L~(-1)組畸形率顯著升高(P0.05)、心率顯著下降(P0.05)。TCCA暴露4和6 h,20.8和41.7 mg·L~(-1)組SOD活性較正常對照組顯著下降(P0.05)。TCCA暴露7 d后,與正常對照組相比,各濃度組斑馬魚幼魚均出現(xiàn)腦和眼部間隙變大及眼部視網(wǎng)膜分層不明顯的病理改變。結(jié)論 TCCA對斑馬魚胚胎發(fā)育有毒性作用,能引起胚胎發(fā)育早期SOD活性下降,造成幼魚視網(wǎng)膜組織結(jié)構(gòu)損傷。
[Abstract]:Objective to study the developmental toxicity of a new disinfectant trichloroisocyanuric acid (TCCA) to zebrafish embryos and young fish. Methods zebrafish embryos were exposed to TCCA 50100150200250 and 300 mg L ~ (-1) for 48 h. The median lethal concentration (LC_ (50). 2) was detected by LC_ (50). 2). The embryo death rate, deformity rate and heart rate were measured at 4872h and 96h after exposure to the culture medium containing TCCA 010.4L ~ (-1) and 41.7 mg L ~ (-1), respectively. The activity of superoxide dismutase (SOD) in embryonic tissues was detected by hydroxylamine method at 2h and 6h after exposure. The histopathological structure of the head of young fish was observed by HE staining at 7 days after exposure. Results the LC_ (50) of zebrafish embryos was 166.9 mg L ~ (-1) after TCCA exposure for 48 h. Compared with the normal control group, the fetal mortality and abnormal rate of zebrafish were significantly increased in TCCA 41.7 mg L ~ (-1) group at 96 h after exposure (P0.05). The mortality rate of TCCA 41.7 mg L ~ (-1) group increased significantly (P0.05), the deformity rate of 20.8 and 41.7mg L ~ (-1) groups increased significantly (P0.05), and the heart rate decreased significantly (P0.05). 48 h after exposure, the deformity rate of TCCA 41.7 mg L ~ (-1) group increased significantly (P0.05), and the heart rate decreased significantly (P0.05). TCCA exposure for 4 and 6 hours). The SOD activity in 20.8 and 41.7 mg L-1 groups was significantly lower than that in the normal control group (P0.05).
