蒙脫石及其制劑吸附力測(cè)定方法研究

【摘要】:目的:對(duì)現(xiàn)行蒙脫石及其制劑質(zhì)量標(biāo)準(zhǔn)中吸附力檢查的原理及方法進(jìn)行深入研究,擬以非毒性試劑三氯六氨合鈷(Ⅲ)替代目前所使用的價(jià)格昂貴且劇毒的硫酸士的寧測(cè)定蒙脫石吸附力。方法:經(jīng)對(duì)蒙脫石吸附力測(cè)定原理的理論論證,優(yōu)化了吸附力測(cè)定方法,利用正交設(shè)計(jì)軟件確定反應(yīng)條件為37℃水浴放置1 h,以紫外-可見(jiàn)分光光度法在474 nm測(cè)定,對(duì)方法的線性、精密度和溶液穩(wěn)定性進(jìn)行了方法學(xué)驗(yàn)證。結(jié)果:經(jīng)對(duì)21家企業(yè)的39批原料與制劑樣品測(cè)定,原料結(jié)果在1.024~1.174 mmol·g~(-1)之間,散劑結(jié)果在1.018~1.238 mmol·g~(-1)之間,分散片結(jié)果在0.894~1.130 mmol·g~(-1)之間,所有樣品結(jié)果與采用硫酸士的寧測(cè)定吸附力的結(jié)果間換算平均系數(shù)為0.43,與理論推導(dǎo)一致,方法可行。結(jié)論:三氯六氨合鈷(Ⅲ)可替代硫酸士的寧作為蒙脫石吸附力測(cè)定的標(biāo)志物,試驗(yàn)安全,無(wú)污染,經(jīng)濟(jì),簡(jiǎn)便。
