高效液相色譜法測定氨基葡萄糖中的乙酰氨基葡萄糖

【摘要】:目的:建立高效液相色譜法測定氨基葡萄糖(Glc N)樣品中乙酰氨基葡萄糖(Glc NAc)。方法:采用氨基柱(250 mm×4.6 mm,5μm),以乙腈-磷酸鹽緩沖液(65∶35,p H 7.5)為流動相,流速1.0 m L·min-1,柱溫30℃,檢測波長195 nm,進(jìn)樣體積20μL。結(jié)果:方法專屬性強(qiáng),Glc NAc與Glc N及各雜質(zhì)分離良好;Glc NAc在1.94~14.70μg·m L-1濃度范圍內(nèi)線性良好(r=0.999 9),準(zhǔn)確度高(回收率99.0%~104.2%,n=3),溶液在24 h內(nèi)穩(wěn)定(RSD為0.69%),精密度(RSD為1.5%~2.2%)和耐用性(RSD為1.6%~8.1%)均良好,靈敏度高(定量限0.98μg·m L-1)。經(jīng)過測定,鹽酸氨基葡萄糖粗品中Glc NAc含量為0.09%~0.18%,鹽酸氨基葡萄糖成品和硫酸氨基葡萄糖氯化鉀成品中未檢出Glc NAc(含量0.0049%)。結(jié)論:本法經(jīng)方法學(xué)驗證,能夠快速、準(zhǔn)確地測定出Glc N中Glc NAc的含量。
