HPLC法測(cè)定小鼠血清中的4-甲基-3-硝基苯甲酸及藥動(dòng)學(xué)研究

【摘要】:目的:建立4-甲基-3-硝基苯甲酸(4-methyl-3-nitro-benzoic acid,MNBA)的HPLC測(cè)定方法,進(jìn)行藥動(dòng)學(xué)研究。方法:用乙腈直接沉淀蛋白提取樣品;采用Intertex C18色譜柱(150 mm×4.6 mm,5μm),以磷酸溶液(p H 0.3)-乙腈(50∶50)為流動(dòng)相,流速1.0 m L·min-1,檢測(cè)波長(zhǎng)230 nm;25℃條件下對(duì)流動(dòng)相、血清樣品預(yù)處理方法進(jìn)行篩選,并對(duì)該方法進(jìn)行方法學(xué)驗(yàn)證。結(jié)果:待測(cè)物質(zhì)分離度良好,質(zhì)量濃度在測(cè)定范圍內(nèi)與峰面積線性關(guān)系良好(r2≥0.999 9),方法精密(日內(nèi)、日間精密度均≤5%)且準(zhǔn)確(低、中、高3種質(zhì)量濃度的準(zhǔn)確度均在95%~110%之間),低、中、高3種質(zhì)量濃度的樣品在4℃冰箱保存10 d,-20℃保存20 d及反復(fù)凍融3次后均保持穩(wěn)定無(wú)降解。小鼠腹腔注射MNBA后在體內(nèi)的代謝過(guò)程符合開(kāi)放一房室模型(w=1/c),其藥代動(dòng)力學(xué)參數(shù)為:T1/2分別為(17.0±3.2)h、(30.5±2.7)h和(80.7±2.0)h;AUC(0-t)分別為(7 801.8±2 360.7)mg·L-1·min-1、(16 589.1±5 383.6)mg·L-1·min-1和(56 898.9±12 153.5)mg·L-1·min-1。結(jié)論:經(jīng)方法學(xué)驗(yàn)證,本法可作為檢測(cè)MNBA的測(cè)定方法;本文測(cè)定的腹腔注射MNBA在小鼠體內(nèi)的藥動(dòng)學(xué)過(guò)程為MNBA的后續(xù)研究提供參考。
